山豆根神经毒性的配伍减毒作用和机制研究 被引量:6

摘要 目的:研究山豆根神经毒性的配伍减毒作用和机制。方法:筛选研究选用具有熄风止痉或平肝潜阳作用的单味药僵蚕、天麻与常用减毒药甘草及所组成复方以不同剂量比例与山豆根配伍,单次灌胃KM小鼠,观察用后神经系统毒性表现,筛选最优减毒处方。减毒机制研究以最优处方配伍灌胃SD大鼠,每天1次,连续2周后测定脑内DA和Ach含量。结果:山豆根与甘草、僵蚕、复方1、复方2配伍时,震颤、异常步态发生率明显降低,有显著性差异(P<0.01或P<0.05),均为1:2配伍比例解毒效果最佳,1:1次之,2:1最弱。综合评介以山豆根与复方2以1:2配伍减毒效果最佳。机制研究表明复方与山豆配伍能降低大鼠脑内Ach浓度(P<0.01),对DA含量无明显影响(P>0.05)。结论:僵蚕、天麻、甘草及其组成的复方与山豆根配伍后能减轻山豆根神经毒性,且有一定的量效关系。以山豆根:复方2(僵蚕+天麻+甘草)按1:2比例配伍时减毒效果最佳。减毒机制可能与复方能调整脑内DA和Ach的比例有关。 Objective: To study the effect and mechanism of combination of attenuated Shandougen neurotoxicity. Methods:We selected the single drug Bombyx Silkworm,Gastrodia elata,and the Compound Radix Glycyrrhizae and the compound compound of Glycyrrhiza uralensis and the Compound Radix Glycyrrhizae in different dosages with the root of KM mice. The toxicity of the posterior nervous system was observed and the best antitoxic prescription was selected. The study of attenuated mechanism was made by adding the best prescription compatibility to SD rats,1 times a day,2 weeks later,the contents of DA and Ach in brain were measured. Results: The incidence of tremor and abnormal gait of Radix Sophorae root,Radix Glycyrrhizae,silkworm,compound 1 and compound 2 were significantly decreased( P〈0. 01 or P〈0. 05),which were the best for 1: 2 compatibility,1: 1 and2: 1 were the weakest. Comprehensive review to Shandougen and compound 2 with 1: 2 attenuated the best compatibility. The mechanism study showed that the combination of compound prescription and Chinese herb could reduce the concentration of Ach in the brain of rats( P〈0. 01),and had no significant effect on the content of DA( P〉0. 05). Conclusion: The combination of Bombyx Silkworm,Gastrodia elata,Glycyrrhiza uralensis and its compound prescription can reduce neurotoxicity of Radix Sophorae Tonkin and have a dose effect relationship. To Shandougen: compound 2( + + silkworm Gastrodia licorice) according to the proportion of 1: 2 and attenuated the best effect. The mechanism of attenuation may be related to the proportion of DA and Ach in the brain.
