摘要
目的:观察黄芪甲苷在高糖条件下对Dll4/Notch-VEGF信号通路主要信号分子Dll4、Notch1、Notch4、VEGF的调控作用,并探讨其抗血管生成的机制。方法:采用30mmol/L葡萄糖培养人脐静脉内皮细胞,设置高糖细胞组(HG)、黄芪甲苷干预组(HQJG+HG)和DAPT组(DAPT+HG)。RT-PCR和Western Blot法检测Dll4、Notch1、Notch4、VEGF的表达。结果:与正常对照组相比,高糖细胞组Dll4、Notch1、VEGF表达上调,Notch4表达下降;采用黄芪甲苷和DAPT干预后可抑制Dll4、Notch1、VEGF表达,上调Notch4表达。结论:黄芪甲苷参与高糖条件下Dll4/Notch-VEGF信号通路的调控,并具有潜在抗血管生成的作用。
Objective: To observe the effect of astragalosideⅣ( AS-Ⅳ) on Dll4/Notch-VEGF signaling pathway,and to analyze the anti-angiogenesis mechanism. Methods: High-glucose( HG) stimulated human umbilical vein endothelial cells( HUVECs) were cultured with 30 mmol/L glucose and set as HG cell model. DAPT and AS-Ⅳ were used in experimental groups,namely DAPT+HG group and AS-IV+HG group respectively. Both expression of mRNA and proteins of Dll4,Notch1,Notch4 and VEGF were recorded. Results: Compared with the normal controlled group,Dll4,Notch1,VEGF expression was upregulated in the HG group with down-expression of Notch4. The results were tured over with intervention of AS-IV and DAPT.Conclusion: AS-IV is involved in the regulation of Dll4/Notch-VEGF signaling pathway under HG stimulation,and might has potential anti-angiogenesis effect.
