摘要
目的:分析干热高温对比湿热高压对附子中6种酯型生物碱含量影响。方法:控制时间与温度,使用湿热高压蒸制与干热高温烘制法炮制附子,使用HPLC检测炮制品内六种生物碱的含量状况。结果:炮制附子使六种生物碱含量产生明显改变,单脂生物碱含量明显升高,双脂生物碱含量消失或者明显下降;乌头碱、次乌头碱及新乌头碱进样量依次在(0.03~3.21,r=0.98)、(0.03~3.10,r=0.98)、(0.03~2.96,r=0.98)间,苯甲酰次乌头原碱、苯甲酰新乌头原碱、苯甲酰乌头原碱进样量依次在(0.03~3.29,r=0.98)、(0.03~3.22,r=0.98)、(0.03~2.98,r=0.98)间,线性关系良好;乌头碱、次乌头碱及新乌头碱回收率RSD依次是0.89%、0.57%、0.98%,苯甲酰次乌头原碱、苯甲酰新乌头原碱、苯甲酰乌头原碱回收率RSD依次是0.99%、1.36%、0.97%。结论:附子炮制采用湿热高压蒸制与干热高压烘制法其方法简便,准确可控,降低毒性同时减少有效成分流失,为炮制附子提供了新的借鉴。
Objective: To analyze the effect of hot and dry high temperature and high temperature on the content of six kinds of ester alkaloids in aconite. Methods: The time and temperature were controlled. The aconite was processed by hot and humid high-pressure steaming and dry-hot high-temperature drying. The contents of six alkaloids in the processed product were detected by HPLC. Results: The results showed that the contents of six alkaloids were obviously changed,the content of mono-lipid alkaloids was significantly increased,and the content of two-lipid alkaloids disappeared or obviously decreased. Aconitine,hypaconitine and mesaconitine in order of( 0. 03 - 3. 21,r = 0. 98),( 0. 03 - 3. 10,r = 0. 98),( 0. 03 - 2. 96,r = 0. 98),Benzoyl aconitine,benzoyl mesaconitine and benzoyl aconitine base were in the order of( 0. 03 - 3. 29,r = 0. 98),( 0. 03 - 3. 22,r = 0. 98)( 0. 03-2. 98,r= 0. 98),the linear relationship was good; The recovery rates of aconitine,hypaconitine and mesaconitine were0. 89%,0. 57% and 0. 98%,respectively. The recovery of the original base RSD of the benzoyl aconitine,the benzoyl mesaconitine and the benzoyl aconitum followed by 0. 99%,1. 36%,0. 97%. Conclusion: The preparation of aconite by using hot and humid high pressure steaming and dry hot high pressure drying method is simple,accurate and controllable,reduce toxicity and reduce the loss of active ingredients,and provide a new reference for the processing of aconite.
