温度对突变型HPV16-L_1蛋白表达的影响

摘要 目的 :探讨温度对突变型 HPV16— L1蛋白表达的影响。方法 :分别在 37℃、2 5℃诱导表达突变型HPV16 - L1蛋白 ,SDS- PAGE电冰比较表达产量。结果 :电泳结果显示 ,降低诱导温度对总蛋白量没有明显影响 ,但目的蛋白量高于常规 37℃下的产量。结论 :诱导表达时适当降低温度可提高目的蛋白表达产量。 Objective:To investigate the effect of temperature on the expression of mutant HPV16-L1 protein.Methods:Mutant HPV16-L1 protein was expressed at 37℃ and 25℃,and the amount of expressed protein was compared by SDS-PAGE.Results:SDS-PAGE analysis showed that the total amount of bacterial protein didnt significantly decrease,however,the amount of recombinant protein increased at 25℃ compared with that of 37℃.Conclusions:Temperature has effect on the expression of mutant HPV16-L1 protein.The amount of recombinant protein would be increasd at 25℃.
