摘要
目的 :研究伴刀豆蛋白 (Concanavalin A,Con A)对红细胞膜机械稳定性的影响 ,以此探讨配体与受体相互作用对红细胞膜力学性质的作用。方法 :利用电子自旋共振技术 (Electron Spin Resonance,ESR)测量经不同浓度 Con A处理后的红细胞膜的 ESR波谱 ,同时还采用剪切流场的方法测量了经Con A作用后的红细胞膜机械稳定性。结果 :经 Con A处理后的红细胞膜蛋白分子构像趋于收缩 ,膜切向机械稳定性明显下降。结论 :Con A作为一种配体与其膜受体 Band3蛋白相结合 ,影响了膜蛋白分子构像 ,进而导致整个红细胞膜机械稳定性发生改变。
Objective:To study the effect of ConA on the mechanical stability of the RBC membrane.Methods:Firstly we measured the RBC membrane treated with the ConA the strong and weak fixation ratio(S/W) in ESR spectrum.Secondly by using the shear flow field to apply shear force on the RBC membrane treated with ConA we measured their hemolysis ration in order to investigate their mechanical stability.Results:The RBC membrane protein conformation was alternated and the RBC membrane mechanical stability declined with the ConA concentration.Conclusion:The interactions between the RBC membrane protein Band3 as receptor and ConA as ligand caused the RBC membrane protein conformation to be changed and resulted in decrease of the membrane mechanical stability.
