细胞分裂素对台湾牛樟腋芽增殖培养的影响 被引量:2

Effects of Cytokinin on the Multiplication and Growth of Axillary Buds of Cinnamomum kanehirae Hayata(Lauraceae)

摘要 为探讨台湾牛樟腋芽增殖系数和丛生芽长势的变化情况,分析细胞分裂素对台湾牛樟腋芽增殖、分化的影响,以2年生台湾牛樟为母树,采用多因素正交试验设计,对腋芽(芽长≥1.0㎝)进行多种激素配比试验。多激素配比试验表明:细胞分裂素与2,4-D组合诱导的台湾牛樟腋芽褐化率较高,腋芽增殖受到抑制;各细胞分裂素对腋芽增殖影响的主次关系为6-BA>ZT>TDZ。6-BA诱导增殖试验表明:增殖系数与腋芽褐化率随6-BA浓度的增加而升高,在6-BA 3.5 mg/L处出现最高峰值。6-BA与ZT组合试验表明:以6-BA 2.0 mg/L和ZT 1.5 mg/L组合的增殖系数最高,腋芽增殖系数3.96,腋芽褐化率整体上明显下降。在台湾牛樟增殖培养阶段,细胞分裂素对其影响较大,其中以6-BA影响最大;6-BA与ZT组合可降低腋芽褐化率,整体上增殖系数提高,芽长势较好。 Cinnamomum kanehirae Hayata was propagated by tissue culture to explore the changes of its multiplication coefficient and growth of cluster buds. Its axillary buds were treated with cytokinin to observe the effect of cytokinin on their differentiation and multiplication. Plants of C. kanehirae Hayata at the age of 2 years old were used as ortets and their axillary buds(bud length ≥1.0 cm) were collected.The axillary buds were treated with combined hormones, and a multi-factorial experiment with orthogonal test was designed. Combined hormones treatment experiment indicated that combination of cytokinin and2,4-D had a higher browning rate and inhibited proliferation of the axillary buds. The effect of cytokinin on the axillary buds of C. kanehirae Hayata was in the order of 6-BA〉 TZ〉TDZ. The 6-BA treatment showed that the multiplication coefficient and the browning rate were increased with the concentration of6-BA, and came to the peak when the axillary buds were treated with 6-BA at the concentration of3.5 mg/L. The 6-BA + ZT treatment indicated that the treatment 6-BA+ZT(2.0 mg/L+1.5 mg/L)gave the highest multiplication coefficient, 3.96 for the axillary buds, and that the browning rate of the axillary buds obviously declined. The cytokinin had a high impact on the axillary buds in the multiplication stage,especially 6-BA which had the greatest impact. The combined 6-Ba and ZT treatment decreased the browning rate and increased the multiplication coefficient, and the cluster buds grew better.
