摘要
目的优选葛根素-黄连素体外抗糖尿病最佳配比。方法采用基线等比增减设计法得到不同配比(10∶0、8∶3、6∶5、3∶8、0∶10),采用DPPH自由基清除、超氧阴离子自由基清除、羟自由基清除、α-葡萄糖苷酶抑制优选体外抗糖尿病最佳配比。结果葛根素-黄连素在不同配比条件下具有清除DPPH自由基、超氧阴离子自由基、羟自由基活性和α-葡萄糖苷酶抑制活性,且葛根素-黄连素(3∶8)时IC_(50)均较小,大小依次为3.71、0.99、4.55、9.65 mg/ml。综合分析得到葛根素-黄连素体外抗糖尿病活性最佳的配比为3∶8。结论葛根素-黄连素在不同配比条件下均具有一定体外抗糖尿病活性,配伍后体外活性优于单个使用,并存在最佳配比。
Objective To optimize the proportion of Puerarin-Berberinein by anti-diabetic activity in vitro. Methods The different proportion are given based on increase-decrease baseline geometric proportion design method, then the optimum proportion of anti-diabetic activity in vitro is established by scavenging DPPH free radicals, reactive oxygen free radicals, scavebging hydroxyl free radicals, α-gucosidase inhibitor. Results Puerarin-Berberinein of the different proportion showed scavenging activity on DPPH free radicals, reactive oxygen free radicals, hydroxyl free radicals and inhibitor activity on α-gucosidase, moreover, the IC50 value is pretty small when the proportion of Puerarin-Berberinein is 3∶8, and the IC50 value is 3.71, 0.99, 4.55, 9.65 mg/ml. Conclusion Puerarin-Berberinein of the different proportion showed anti-diabetic activity in vitro, the activity in vitro of compatibillty is superior to single, and had optimal proportion.
