摘要
比较了分光光度法和高效液相色谱(HPLC)法测定地表水中叶绿素a的结果。研究表明:两种方法的精密度无显著差异;叶绿素a标准样品测定结果也无显著差异;在测定地表水样品时,分光光度法测定结果高于HPLC法,差异主要由664 nm波长下有较强吸收的物质造成。分光光度法测定结果偏高但操作简单,HPLC法测定结果精准但操作复杂,分光光度法在水质监测工作中更加具有优势。
The determination results of chlorophyll a in surface water were compared between spectrophotometry and high performance liquid chromatography(HPLC). The study showed that there was no significant difference in the precision of the two methods,as well as the chlorophyll a standard determination results. When it came to surface water samples,the results of spectrophotometry were higher than those of HPLC. The difference was mainly made by substances with strong absorption at 664 nm. Spectrophotometry was not accurate but easy to operate,HPLC was accurate but complicated,spectrophotometry had more advantage in monitoring water quality.
