摘要
目的探讨M型钾离子通道开放剂对心肺复苏大鼠全脑损伤的保护作用。方法取健康Wistar大鼠60只,随机分为假手术组、心脏骤停复苏组及M型钾离子通道开放剂干预组,每组20只。按心肺复苏后自主循环恢复6、12、24、48、72 h,每组将4只大鼠处死,取脑组织,检测一氧化氮、神经元特异性烯醇化酶、高迁移率蛋白B1、脑组织含水量。处死前,给予大鼠神经功能缺损评分。结果心脏骤停复苏组和M型钾离子通道开放剂干预组随自主循环恢复时间延长,一氧化氮、神经元特异性烯纯化酶、高迁移率蛋白B1、脑组织含水量呈先增加后降低态势,最高点在24 h。神经功能缺损评分呈增加态势。心脏骤停复苏组在自主循环恢复的不同时间点(6 h、12 h、24 h、48 h、72 h)一氧化氮、神经元特异性烯纯化酶、高迁移率蛋白B1、脑组织含水量高于假手术组和M型钾离子通道开放剂干预组。心脏骤停复苏组在自主循环恢复的不同时间点(6 h、12 h、24 h、48 h、72 h)神经功能缺损评分低于假手术组和M型钾离子通道开放剂干预组。M型钾离子通道开放剂干预组在自主循环恢复的不同时间点(6 h、12 h、24 h、48 h、72 h)一氧化氮、神经元特异性烯纯化酶、高迁移率蛋白B1、脑组织含水量高于假手术组。M型钾离子通道开放剂干预组在自主循环恢复的不同时间点(6 h、12 h、24 h、48 h、72 h)神经功能缺损评分低于假手术组,差异均有统计学意义(P<0.05)。结论 M型钾离子通道开放剂可减轻心肺复苏大鼠的全脑损伤。
Objective To investigate the protective effects of M type potassium channel opener on brain injury of rats after cardiopulmonary resuscitation. Methods Sixty healthy Wistar rats were randomly divided into sham operation group,cardiac arrest resuscitation group,M type potassium channel opener intervention group,with 20 rats in each group. According to spontaneous circulation recovery time after cardiopulmonary resuscitation at 6 h,12 h,24 h,48 h,72 h,the 4 rats in each group were sacrificed,and brain tissues were collected,the levels of nitric oxide,neuron specific enolase,high mobility protein B1,brain tissue water content were detected. Moreover the neurological deficit scoring was performed before the rats were sacrificed. Results The spontaneous circulation recovery time was lengthened in cardiac arrest resuscitation group and M type potassium channel opener intervention group,and the levels of nitric oxide,neuron specific enolase,high mobility protein B1,brain tissue water content were increased at first and then decreased,with highest point being at 24 h. The neurological deficit scores showed an increasing trend. The levels of nitric oxide,neuron specific enolase,high mobility protein B1 and brain tissue water content at different time points( 6 h,12 h,24 h,48 h,72 h) after cardiopulmonary resuscitation in cardiac arrest resuscitation group were higher than those in sham operation group and M type potassium channel opener intervention group.Moreover the neurological deficit scores at different time points( 6 h,12 h,24 h,48 h,72 h) after cardiopulmonary resuscitation in cardiac arrest resuscitation group were lower than those in sham operation group and M type potassium channel opener intervention group. In addition the levels of nitric oxide,neuron specific enolase,high mobility protein B1 and brain tissue water content at different time points( 6 h,12 h,24 h,48 h,72 h) after cardiopulmonary resuscitation in M type potassium channel opener intervention group were higher than those in sham operation group,moreover,the neurological deficit scores at different time points( 6 h,12 h,24 h,48 h,72 h) after cardiopulmonary resuscitation in M type potassium channel opener intervention group were lower than those in sham operation group,and these differences were statistically significant( P〈 0. 05).Conclusion The M type potassium channel opener can relieve whole brain injury of rats after cardiopulmonary resuscitation.
