河北省区域试验大豆品系指纹图谱构建遗传相似性分析及纯度鉴定 被引量:3

DNA Fingerprinting, Genetic Similarity and Purity Identification Analysis of Soybeans Lines Attending Regional Tests of Hebei Province

摘要 通过鉴定河北省区域试验大豆品系的纯度、构建参试大豆品系的指纹图谱、分析品系间的遗传关系,可有效指导河北省大豆新品种的选育和推广。以参加2016年河北省区域试验的21个大豆品系为材料,用均匀分布在20条染色体上的62对SSR标记进行分析。结果表明:3对SSR引物(Satt216、Satt175和Satt125)构建的指纹图谱可以将21个大豆品系完全区分开;采用非加权类平均法进行聚类分析,21个大豆品系的遗传相似系数范围为0~0.82,平均0.63。以遗传相似系数0.64为阈值可以将21份品系分为3大类群,其中,第二类群与第三类群品系间的遗传相似系数范围为0.61~0.82。通过筛选的2对SSR引物(Satt216、Satt114)检测到大豆品系纯度分布范围为85%~100%,平均纯度为97%。21份参试品系的遗传相似系数较高,品系间遗传变异较小,亲本材料的遗传背景比较相近,在一定程度上导致大豆遗传基础比较狭窄。 The purity identification, DNA fingerprinting construction, and genetic relation analysis of soybean lines in regional test of Hebei could guide the breeding and population of new soybean verities. 62 pairs SSR markers distributed in 20 chromosomes of soybean were used to identify 21 soybean lines in regional test of2016 in Hebei Province. The results showed that 21 soybean lines were distinguished from each other by 3 pairs SSR markers(Satt216,Satt175 and Satt125). The genetic similarity(GS) coefficients of 21 soybean lines by unweighted pair group mathematics average(UPGMA)clustering were 0-0.82,averaging 0.63. When GS was0.64, all lines could be ascribed into 3 groups. The GS between second and third group were 0.61-0.82.The purity of soybean lines was detected by 2 pairs of SSR markers(Satt216 and Satt114), the purity distribution ranged from 85% to 100%, and the average purity was 97%. The GS of 21 soybean lines was high. There was little variation among lines. The genetic background of parents was similar, which resulted in narrow genetic basis of soybean to a certain degree.
