绝经后骨质疏松症患者基因表达谱的改变 被引量:1

摘要 目的利用基因芯片公共数据库中RNA标本芯片数据,分析绝经后骨密度降低程度不同的骨质疏松症患者外周血基因表达谱及分子网络变化。方法从基因芯片公共数据库(Gene Expression Omnibus,GEO)中下载美国奥马哈20名绝经后妇女的外周血RNA标本基因芯片数据,利用基因云、基因大数据分析(Gene-Cloud of Biotechnology Informs,GCBI)实验平台、Gen Clip2.0、Sytoscape 3.5.1等软件,筛选差异表达基因。结果差异基因主要涉及MAPK调节、细胞内信号转导的调控、细胞运动、免疫效应等生物学功能。基因网络中以MAPK3、ARRB2、STK11、USP9X、MYO1C、GTF2H1等基因degree值较大,基因MAPK3、ARRB2、STK11、MYO1C、GTF2H1在不同骨密度人群中表达差异有统计学意义(P<0.05),其中基因MAPK3、ARRB2、STK11和MYO1C诊断价值有统计学意义(P<0.05)。结论 MAPK3、ARRB2、STK11和MYO1C基因为蛋白网络核心节点。MAKP3和ARRB2基因与骨质疏松的发生关系密切。 Objective To analyze the biological network regulation and key proteins of peripheral blood gene expression microarray data of postmenopausal patients with different bone mineral densities. Methods The data of RNA microarray in peripheral blood samples of 20 postmenopausal women from Omaha, Omaha, USA were dow nloaded from Gene Expression Omnibus (GEO) database. Gene-Cloud of Biotechnology Informs (GCBI) Platform, GenClip2.0, Sytoscape 3.5.1 and other software were used to screen differentially expressed genes, explore the protein-protein interaction networks and biological pathways of differentially expressed genes, and elucidate the molecular mechanism of osteoporosis development from the perspective of transcriptome. Results The differential genes mainly involved in the regulation of MAPK, the regulation of intracellular signal transduction, cell motility, immune effects and other biological functions. Genes such as MAPK3, ARRB2, STK11, USP9X, MYO1C and GTF2H1 had higher gene value. Among them, the gene expression of MAPK3, ARRB2, STK11, MYO1C and GTF2H1 in different BMD population was statistically significant ( P 〈0.05), while the diagnostic value of MAPK3, ARRB2, STK11 and MYO1C were statistically significant ( P 〈0.05). Conclusion MAPK3, ARRB2 , STK11 and MYO1C are the core nodes of protein network, which are highly expressed in high BMD group. The functions of MAPK3, ARRB2, STK11 and MYO1C were mainly involved in the biological functions of MAPK regulation, regulation of intracellular signal transduction, cell motility and immune response.
