黏着斑激酶抑制剂TAE226对人舌鳞状细胞癌Tca8113细胞的体外迁移和侵袭的影响

摘要 目的研究黏着斑激酶(FAK)抑制剂TAE226对人舌鳞状细胞癌Tca8113细胞的体外迁移和侵袭能力的影响。方法在Transwell迁移实验和Transwell侵袭实验中,通过计数TAE226作用于Transwell上室细胞24 h后的细胞穿膜贴壁数计算TAE226对细胞的迁移和侵袭的影响;在划痕实验中,通过划痕后加入不同浓度的TAE226(0、1、5、10μmol/L)作用于细胞,并于划痕后的0、12以及24 h测量划痕宽度的变化,计算细胞的迁移率。结果 (1)Transwell迁移实验结果:与对照组相比,实验组的穿膜细胞数减少,随着TAE226浓度的升高,Tca8113穿膜细胞数分别为:对照组(216.80±10.64)个;1μmol/L实验组(149.00±7.91)个、5μmol/L实验组(127.00±5.57)个、10μmol/L实验组(104.40±6.80)个、20μmol/L实验组(61.40±7.76)个,差异有统计学意义(F=264.339,P<0.05)。(2)划痕实验结果:与对照组相比,不同浓度的实验组均能抑制细胞的迁移,实验组中浓度为1和5μmol/L的TAE226对细胞的迁移抑制作用存在剂量依赖性(P<0.05),而5和10μmol/L的实验组对细胞的迁移抑制作用比较,差异无统计学意义(P>0.05)。TAE226对各组的迁移抑制作用均存在时间依赖性(P<0.05)。(3)Transwell侵袭实验结果:不同浓度的TAE226(1、5、10、20μmol/L)侵袭率分别为76.72%、63.36%、43.10%、22.84%,随着TAE226浓度的升高,对细胞的侵袭抑制作用增强(P<0.05)。结论 TAE226可以有效抑制Tca8113细胞的迁移及侵袭。 Objective To study the effects of a novel focal adhesion kinase(FAK) inhibitor TAE226 on the migration and invasion of human oral tongue squamous cell carcinoma cell line Tca8113 in vitro. Methods In the transwell migration experiment and transwell invasion experiment, the effects of TAE226 on cell migration and invasion were calculated by counting the number of cells which sticking through the membrane after 24 hours of TAE226 acting on transwell. In the scratch experiment, the cells were treated with the different concentrations of TAE226(0, 1, 5, 10 μmol/L) , and the rate of the migration was worked out by measuring the change of the length of scratch width after it was scratched for 0, 12, 24 h. Results ①The result of transwell migration experiment: compared with control group, the number of perforating cells in the experimental group was significantly reduced. With the increase of the concentration of TAE226, the number of perforating cells in the control group was (216.80±10.64),1 μmol/L experiment group was (149.00±7.91) and 5 μmol/L experimental group was (127.00±5.57) and 10 μmol/L experimental group was (104.40±6.80) and 20 μmol/L experimental group was (61.40±7.76) , and the difference was statistically significant ( F =264.339, P 〈0.05).②The result of scratch test: compared with control group, experimental group can inhibit the migration of the cells with different concentration of TAE226 in scratch healing assay. The group of 1 μmol/L and 5 μmol/L in a dose dependent manner( P 〈0.05), and the different inhibitory effects of the group of 5 μmol/L and 10 μmol/L were not statistically significant( P 〉0.05). The migration inhibition rates were time dependent( P 〈0.05). ③The result of transwell invasion experiment was that the invasion rates after co-cultured of different concentrations of TAE226(1, 5, 10, 20 μmol/L) were 76.72%, 63.36%, 43.1%, 22.84%. With the increase of TAE22 centration, the inhibition of cell invasion was enhanced( P 〈0.05). Conclusion TAE226 can effectively inhibit the migration and invasion of human tongue carcinoma cells line Tca8113.
