摘要
目的血红素加氧酶1(HO-1)在间歇性低氧对大鼠心肌缺血再灌注(I/R)损伤的影响。方法 54只大鼠分为随机3组:假手术组(n=18)、缺血再灌注组(n=18)、间歇性低氧+缺血再灌注组(n=18)采用球囊结扎冠状动脉方法制作心肌缺血再灌注动物模型,将老鼠暴露在低氧循环制作间歇性低氧动物模型,实验处理8周后,通过超声心动图测量左心室舒张期末容积、左心室收缩期末容积、左心室射血分数通过Western blot法检测HO-1蛋白表达。结果假手术组、缺血再灌注组、间歇性低氧+缺血再灌注组左心室舒张期末容积、左心室收缩期末容积、左心室射血分数分别为(426±3.2)μL、(182±4.1)μL、(57±3.9)%,(782±1.6)μL、(547±3.5)μL、(29±1.6)%,(518±4.7)μL、(392±2.8)μL、(46±5.3)%,组间比较均有统计学差异(P<0.05);假手术组、缺血再灌注组及间歇性低氧+缺血再灌注组HO-1蛋白表达分别为0.392±3.176、0.217±1.326、0.982±4.538(P<0.05)。结论 HO-1在间歇性低氧状态下,对大鼠缺血再灌注损伤具有保护作用。
Objective To research the effects of heme oxygenase-1 on heart ischemic/reperfusion myocardial damage in Intermittent Hypoxia. Methods Fifty-four ats were randomly divided into 3 groups sham-operated group(n=18); myocardial ischemia reperfusion group(I/R)(n=18), I/R+ Intermittent Hypoxia group(n=18). We established the IH mode through intermittent hypoxia cycles, I/R model was established by ligating the left anterior descending coronary artery. We assessed the LV end-diastolic volume (LVEDV), LV end-systolic volume (LVESV), LV ejection fraction (LVEF) of the different groups by echocardiography after 8 weeks, and the expressions of HO-1 were determined by Westen blot. Results The LVEDV, LVESV, LVEF of sham-operated group, I/R group, IH+I/R group was(426±3.2) μL, (182±4.1) μL, (57±3.9)%; (782±1.6) μL, (547±3.5) μL, (29±1.6)%; (518±4.7) μL, (392±2.8) μL, (46±5.3)%; there was signifcant difference between the groups (P〈0.05); compared with sham-operated group, I/R group, IH+I/R group was 0.392±3.176, 0.217±1.326, 0.982±4.538, there was signifcant difference between the groups (P〈0.05). Conclusion HO-1 has a cadioprotective effect on heart ischemic/reperfusion damage in intermittent hypoxia.
