加味地黄饮子对衰老小鼠肾脏组织P21、P53蛋白及其mRNA表达的影响 被引量:8

Effect of Jiawei Dihuang Yinzi on Protein and mRNA Expression of P21 and P53 in Kidney Tissue in Senescent Mice

摘要 目的观察加味地黄饮子对衰老小鼠肾脏组织P21、P53蛋白及其mRNA表达的影响。方法选取2月龄小鼠10只作为青年组,将18月龄小鼠30只随机分为老年组、维生素E组和加味地黄饮子组。青年组、老年组给予生理盐水灌胃,维生素E组、加味地黄饮子组给予对应药物灌胃。干预60d后,观察各组小鼠外观特征及肾脏指数改变,检测血清肌酐(serum creatinine,SCr)、血尿素氮(blood urea nitrogen,BUN),观察各组肾脏病理学改变,检测超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)水平,RT-PCR法检测P21、P53mRNA表达,Western-blot法检测P21、P53蛋白的表达。结果与老年组比较,加味地黄饮子组小鼠运动活跃,反应敏捷,SOD、GSH-Px水平显著升高,肾脏指数、SCr、BUN、MDA水平显著降低,P21、P53蛋白及其mRNA表达水平明显降低,差异均有统计学意义(P<0.05);加味地黄饮子组小鼠肾小球硬化、肾小管萎缩、系膜基质增厚、炎性细胞浸润均较老年组明显减轻。结论加味地黄饮子可通过干预P21、P53及其mRNA表达延缓小鼠肾脏衰老。 Objective To investigate the effect of Jiawei Dihuang Yinzi on the protein and mRNA expression of P21 and P53 in kidney tissue in senescent mice. Methods A total of 10 mice aged 2 months were selected as young group, and 30 mice aged 18 months were randomly divided into senescent group, vitamin E group, and Jiawei Dihuang Yinzi group. The mice in the young group and the senescent group were given normal saline by gavage, and those in the vitamin E group and the Jiawei Dihuang Yinzi group were given vitamin E or Jiawei Dihuang Yinzi by gavage. After intervention for 60 days, the changes in appearance and kidney index were evaluated; serum creatinine (SCr) and blood urea nitrogen (BUN) were measured; renal pathological changes were assessed; superoxide dismutase (SOD), malondialdehyde (MDA), and glutathione peroxidase (GSH-Px) were measured; RT-PCR and Western blot were used to measure the mRNA and protein expression of P21 and P53. Results Compared with those in the senescent group, the mice in the Jiawei Dihuang Yinzi group were more active and reacted more quickly; compared with the senescent group, the Jiawei Dihuang Yinzi group had significant increases in the levels of SOD and GSH-Px and significant reductions in kidney index and the levels of SCr, BUN, and MDA, as well as significant reductions in the protein and mRNA expression of P21 and P53 (P〈0.05). Compared with the senescent group, the Jiawei Dihuang Yinzi group had marked alleviation of glomerular sclerosis, tubular atrophy, thickening of the mesangial matrix, and inflammatory cell infiltration. Conclusion Jiawei Dihuang Yinzi may delay the aging of the kidney in mice by regulating the protein and mRNA expression of P21 and P53.
