摘要
目的观察900 MHz手机频率电磁辐射对雄性大鼠睾丸组织氧化损伤和Nrf2蛋白表达的影响。方法30只SD大鼠分为正常组、辐射2 h组和辐射4 h组(n=10)。正常组辐射0 h,辐射2 h组和辐射4 h组每天分别暴露于900 MHz电磁辐射(370μW/cm^2)辐射2 h和4 h,连续30 d。光镜观察各组大鼠睾丸组织形态学变化,比色法检测睾丸组织超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、丙二醛(MDA)变化,免疫组化和Western blot检测核因子相关因子-2(Nrf2)的蛋白表达。结果与正常组比较,辐射组大鼠睾丸生精小管结构稍见松散;大鼠睾丸组织MDA含量均明显增加,SOD活性、GSH含量显著降低(P<0.05);辐射组大鼠睾丸组织Nrf2蛋白表达明显减少(P<0.05)。与辐射2 h组比较,辐射4 h组管腔内成熟精子减少;大鼠睾丸MDA含量均明显增加,SOD活性和GSH含量显著降低(P<0.05);睾丸组织Nrf2蛋白表达明显减少(P<0.05)。结论 900 MHz频率电磁辐射可加重大鼠睾丸组织氧化损伤,可能与减弱Nrf2在睾丸组织的表达有关,并且电磁辐射与氧化损伤有时效关系。
Objective To observe the effect of 900 MHz electromagnetic radiation( EMR) of cellphone on oxidative damage and expression of Nrf2 protein in testis tissues of male rats. Methods Thirty adult male SD rats were divided into normal group( n = 10),2 h-radiation group( n = 10) and 4 h-radiation group( n = 10),with 10 rats in each group. The rats in normal group were not exposed to EMR,And the rats in 2 h-radiation group and 4 h-radiation group were exposed to 900 MHz EMR( 370μW/cm-2) 2 h and 4 h every day,respectively,for 30 consecutive days. The morphological changes of testicular tissue of rats were observed,and the changes of MDA and GSH in testes tissues were detected by colorimetry,moreover,the protein expression levels of nuclear factor related factor were measured by immunohistochemisty and Western Blot. Results As compared with those in normal group,the testicular seminiferous tubules of the rats in radiation groups became loose slightly,and the content of MDA in testicular tissue of rats was significantly increased,while SOD activity and GSH content were significantly decreased( P〈0. 05). The expression levels of Nrf2 protein in testicular tissue of the rats in radiation groups were significantly decreased( P〈0. 05). As compared with those in 2 h-radiation group,the spermatozoa in lumens in4 h-radiation group were reduced,and the content of MDA in rat testes was increased significantly,however,the SOD activity and GSH content were significantly decreased( P〈0. 05),and the expression levels of Nrf2 protein in testicular tissue were significantly decreased( P〈0. 05). Conclusion The 900 MHz electromagnetic radiation can aggravate the oxidative damage of testicular tissue in rats,which may be related to the weakening of Nrf2 protein expression in testis. Moreover EMR induced oxidative damage has a time-effect relationship.
