摘要
目的:检测非编码微小RNA135b在宫颈癌中的表达,并初步探讨其可能的作用机制。方法:Realtime PCR检测19例宫颈癌组织和癌旁组织中miR-135b-5p的表达量,并和患者年龄、细胞分化程度、临床分期、淋巴结转移的相关性进行分析;同时通过软件预测miR-135b-5p的目的基因KLF4,Western Blot检测靶基因蛋白的表达变化。结果:Real-time PCR结果显示,和癌旁组织相比,宫颈癌中miR-135b-5p的表达量明显增高,差异有统计学意义(P<0.05);而且和宫颈癌患者的细胞分化程度、临床分期、淋巴结转移呈正相关(P<0.05);Western Blot检测结果显示KLF4在宫颈癌组织中表达量较癌旁组织减低(P<0.05)。结论:miR-135b-5p能够促进宫颈癌的发生和发展,可能和抑制KLF4的表达有关。
Objective:To detect the role of non-coding microRNA135b in cervical cancer and to explore its possible mechanism.Methods:Real-time PCR was used to detect the expression of miR-135b-5p in cervical cancer tissues and adjacent tissues.The correlation between miR-135b-5p and clinical stage,lymph node metastasis and depth of cancer invasion was analyzed.The expression of target gene protein was detected by KLF4 and Western Blot,the target gene of miR-135b-5p.Results:Real-time PCR results showed that the expression of miR-135b-5p in cervical cancer was significantly higher than that in paracancerous tissues,and the difference was statistically significant.The clinical stage of cervical cancer patients and the localized lymph node metastasis and the depth of invasion was positively correlated.Western Blot test showed that KLF4 expression in cervical cancer was lower than that in adjacent tissues.Conclusion:miR-135b-5p can promote the occurrence and development of cervical cancer,which may be related to the inhibition of KLF4 expression.
