陕产细叶益母草中总生物碱的提取工艺研究 被引量:2

摘要 目的:优选陕产细叶益母草中总生物碱的提取工艺。方法:利用响应曲面法,在单因素实验的基础上,以超声时间、乙醇浓度、料液比为自变量,采用雷氏盐比色法,以益母草生物碱的得率为响应值,应用中心组合设计实验方法,研究各自变量及其交互作用对生物碱得率的影响,建立二次多项回归方程的数学模型。结果:料液比对益母草生物碱得率的影响比较显著。结论:最终优选的超声法提取益母草生物碱工艺为:在超声功率为180 W,温度为40℃,0.5%盐酸的酸性条件下,优化后的料液比为1∶18,提取时间为40 min,乙醇浓度为74%,在该条件下,益母草生物碱得率的理论值为0.195 27%。 Objective:To optimize the process conditions for the total alkaloid of Leonurus artemisia originateed from shanxi. Methods: Response surface methodology was used on the basis of single factor experiment. Ultrasonic time, ethanol concentration, solid - liquid ratio were taken as independent variables, the alkaloids yield of Leonurus artemisia determined by the Ammoniun Reineckate - Differential - Colorimetry method was as theresponse value, effect of interaction of each variable on the yield of alkaloids was investigated with the application of central composite experimental design method, and the mathematical model of two multinomial regression equation was established. Results: Solid - liquid ratio had obvious effect on the extraction rate of alkaloid. Conclusion: The final optimization process for Leonurus artemisia is as follows: ultrasonic power is 180 Wat the temperature of 40℃ under 0. 5% hydrochloric acid conditions, the optimized solid - liquid ratio is 1∶ 18, extraction time is 40 min, and ethanol concentration is 74%, under these condition, the yield of alkaloid of Leonurus artemisia is 0. 195 27%.
