加味乌梅丸联合吉西他滨对胰腺癌移植瘤的抑制及细胞凋亡的实验研究

Experimental study on inhibition and apoptosis of transplanted tumor of pancreatic cancer with Jiawei Wumei pill combined with gemcitabine

目的探讨加味乌梅丸对胰腺癌移植瘤的抑制作用及其作用机理。方法建立裸鼠人胰腺癌细胞株SW1990皮下移植瘤模型,将40只裸鼠按体重随机分为4组,模型组、中药组、化疗组、中药+化疗组,记录各组移植瘤的生长情况,计算肿瘤体积,绘制生长曲线;21天后处死裸鼠,称取瘤重,计算抑瘤率。HE染色下进行病理学观察,TUNEL法检测肿瘤组织的细胞凋亡情况。结果 (1)中药组、化疗组和中药+化疗组的平均瘤重和瘤体积均小于模型组,差异均有统计学意义(P<0.05);三组抑瘤率分别为24.36%、48.59%、63.88%,其中中药+化疗组的实际抑瘤率(63.88%)大于两组的理论相加抑瘤率(61.11%);中药组、化疗组和中药+化疗组的肿瘤坏死率分别为(16.89±4.9)、(17.43±8.04)、(20.98±2.26),与模型组(7.67±3.69)相比,差异均有统计学意义(P<0.05);中药组、化疗组和中药+化疗组肿瘤组织中凋亡细胞均数分别为(61.29±12.84)、(65.28±11.94)、(100.83±27.12),明显大于模型组的(40.57±12.71);差异均有统计学意义(P<0.05);其中中药+化疗组与化疗组相比,其差异亦有统计学意义(P<0.05)。结论加味乌梅丸可抑制胰腺癌移植瘤的生长,与吉西他滨联合有协同抑制作用,其抑瘤机制主要是促进肿瘤细胞的凋亡。

Objective To investigate the inhibitory effect of Jiawei Wumei Pill on pancreatic cancer transplantable tumor and its mechanism. Methods Subcutaneously transplanted tumor model of human pancreatic cancer cell line SW1990 in nude mouse was established,40 nude mice were randomly divided into model group,Chinese medicine group,chemotherapy group,Chinese medicine ＋ chemotherapy group,the growth of tumor in each group was observed and recorded,and the volume of tumor was calculated,At the same time,the tumor growth curve was drawn. After treatment of 21 days,the mice were sacrificed. The weight of tumor was evaluated. HE staining was used to observe the pathological changes,and the apoptosis of tumor tissues was detected by TUNEL. Results（ 1） The average tumor weight and tumor volume in the 3 treatment groups were smaller than the model group,the differences were statistically significant（ P〈0. 05）; The tumor inhibition rate in three groups were 24. 36% 、48. 59% 、63. 88%,The actual inhibition rate in Chinese medicine ＋ chemotherapy group was 63. 88%,which was higher than the inhibition rate by theoretical addition（ 61. 11%）; The tumor necrosis rate in three groups were（ 16. 89 ± 4. 9） 、（ 17. 43 ± 8. 04） 、（ 20. 98 ± 2.26）,compared with model group,tumor necrosis rate in chemotherapy group was（ 7. 67 ± 3. 69）,the differences were statistically significant（ P〈0. 05）. The mean apoptotic cell number in three groups were（ 61. 29± 12. 84）,（ 65. 28 ± 11. 94） and（ 100. 83 ± 27. 12）,significantly higher than the model group（ 40. 57 ± 12.71）,the differences were statistically significant（ P〈0. 05）; Compared with the chemotherapy group,the difference was statistically significant（ P〈0. 05） in the Chinese medicine ＋ chemotherapy group. Conclusion Jiawei Wumei Pill can inhibit the growth of pancreatic cancer transplantable tumor,and gemcitabine have synergistic inhibitory effect,the inhibition mechanism is mainly by promoting apoptosis of tumor cells.
