摘要
目的观察养阴润肠方不同剂量对(slow transit constipation,STC)模型(interstitial cells of cajal,ICC)的影响。方法随机将SD大鼠分为正常组、模型组、养阴润肠方低剂量组、养阴润肠方中剂量组、养阴润肠方高剂量组。在药物干预的第2、4、6周后进行结肠内存留粪便粒数、碳末推进率、结肠ICC细胞形态学观察。结果整体实验证实养阴润肠方在给药第4周时,为目前最佳给药时间,因此观察养阴润肠方在给药4周后的各项指标。观察粪便含水量发现与模型组相比较,养阴润肠方低剂量组、中剂量组及高剂量组均能提高粪便含水量,差异具有统计学意义(P<0.01),养阴润肠方中剂量组、高剂量组之间未见明显差异,无统计学意义(P>0.05),养阴润肠方低剂量组在提高粪便含水量的能力方面弱于中剂量组及高剂量组,差异具有统计学意义(P<0.05)。在粪便残留数及碳墨推进率方面,养阴润肠方中剂量组及高剂量组与模型组比较,均具有明显改善,差异有显著统计学意义(P<0.01),但中剂量组、高剂量组之间未见明显差异,而低剂量组无明显改善,差异无统计学意义(P>0.05)。结论养阴润肠方中剂量具有确切的促进STC大鼠模型结肠传输、修复ICC细胞的功能,不需增加或减少剂量。
Objective To observe the difference dose of Yangyin Runchang prescription on slow transit constipation( STC) model ICC activation state. Methods SD rats were randomly divided into normal group,model group,Yangyin Runchang low dose,middle dose and high dose groups. After the 2^nd,4^th,and 6^th week of drug intervention,the number of stool feces in the colon,the carbon advancing rate,and the morphology of colonic ICC cells were observed. Results The overall experiment confirmed that the Yangyin Runchang prescription was the best time for administration at the 4^th week,so the indicators of Yangyin Runchang prescription were observed after 4 weeks of administration. The observation on the water content of feces found that compared with the model group,the low-dose,middle-dose and high-dose groups of Yangyin Runchang prescription could increase the water content of feces,and the difference was statistically significant( P〈0. 01). There was no significant difference between the middle dose group and the high dose group( P〉0. 05). The low dose group of Yangyin Runchang prescription was weaker than the middle dose group and the high dose group in increasing the water content of feces. The difference was statistically significant( P〈0. 05). In terms of the number of fecal residues and the rate of carbon ink advancement,the middle dose group and the high dose group were significantly improved compared with the model group,and the difference was statistically significant( P〈0. 01). But there was no significant difference between the middle-dose and high-dose groups,and there was no significant improvement in the low-dose group without statistical significance( P〉0. 05). Conclusion Yangyin Runchang prescription has the function of promoting colonic transmission and repairing ICC cells in STC rat model without needs of increasing or decreasing the dose.
