摘要 目的探讨多因素诱导足细胞损伤情况下,新骨架蛋白分子transgelin的表达改变,揭示新确定的足细胞骨架蛋白分子transgelin的生物学效应。方法血管紧张素Ⅱ(AngⅡ)、阿霉素(Adriamycin)诱导足细胞损伤,间接免疫荧光染色检测AngⅡ、Adriamycin诱导足细胞损伤下微丝骨架纤维状肌动蛋白(F-actin)的分布。蛋白印迹法(Western blotting)检测transgelin的蛋白表达。结果 AngⅡ、Adriamycin可诱导微丝骨架F-actin分布改变。Transgelin与F-actin具有共定位关系。AngⅡ、Adriamycin可诱导足细胞transgelin表达增加,差异具有统计学意义(P<0.05)。结论在Ang II、Adriamycin致体外培养的足细胞损伤的过程中,transgelin表达上调,提示transgelin参与了多种因素引起的足细胞损伤过程。 Objective To investigate the role of a novel skeletal protein transgelin in podocytes in vitro. Methods Podocytes were treated with Angiotensin( Ang Ⅱ) and Adriamycin for indirect immunofluorescence staining assessment of Ang Ⅱ or Adriamycin-induced microfilament cytoskeleton changes. Level of transgelin expression was assessed by Western blot. Results The data showed that Ang Ⅱ and Adriamycin induced expression and re-organization of the microfilament skeleton F-actin in podocytes. Transgelin expression was induced by Ang Ⅱ and Adriamycin treatment and co-localized with F-actin in podocytes( P〈0. 05). Conclusion Transgelin expression is up-regulated by Ang Ⅱ and Adriamycin treatment and is associated with Ang Ⅱ and Adriamycin treatment-induced podocyte injury in vitro,suggesting that transgelin may be involved in Ang Ⅱ and Adriamycin treatment-induced podocyte damage.
