羊耳菊水提物对重症肺炎大鼠血清炎症因子的影响及机制初探 被引量:6

摘要 目的探讨羊耳菊水提物对重症肺炎(SP)大鼠血清炎症因子的影响,并初步探讨其机制。方法取45只大鼠建立SP模型,将建模成功的大鼠随机分为模型组,低、中、高剂量组,另外10只大鼠为对照组。低、中、高剂量组按照20 ml/kg灌胃,保证羊耳菊生药摄入量为6 g/kg、12 g/kg、24 g/kg,模型组和对照组灌胃等量蒸馏水,连续6 d。对比灌胃2 d、4 d、6 d时血清炎症因子水平;对比肺组织中Toll样受体2(TLR2)、核转录因子κB(NF-κB)p65、髓样细胞分化因子88(My D88)mRNA及蛋白相对表达量。结果共40只大鼠建模成功。血清白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)水平、TLR2、My D88、NF-κB p65 mRNA和蛋白相对表达量组间比较,对照组最低、中剂量组次之、低、高剂量组稍高、模型组最高,除低、高剂量组比较差异不显著(P>0.05),其他每两组间比较差异均有显著性(P<0.05);血清TNF-α、IL-6、IL-1β、TLR2水平组内比较,3剂量组均随时间延长显著降低(P<0.05),模型组随时间延长显著升高(P<0.05),对照组随时间延长变化不显著(P>0.05)。结论羊耳菊水提物可改善SP大鼠炎症反应,其中12 g/kg的羊耳菊水提物改善作用最佳,可能与抑制TLR2/My D88/NF-κB信号通路有关。 Objective To investigate the effect of Inula Cappa aqueous extract on serum inflammatory factors in rats with severe pneumonia( SP) and to explore its mechanism. Methods 45 rats were selected to establish SP model. The successful SP model rats were divided into model group,low,middle and high dose groups,and the other 10 rats were selected as control group. The low,middle and high dose groups were fed with 20 ml/kg,which ensured that the intakes of the Inula Cappa crude drug were 6 g/kg,12 g/kg and 24 g/kg respectively. The model group and the control group were fed with equal amount of distilled water for 6 days. The serum inflammatory factors were compared after 2,4 and 6 days of intragastric administration. The mRNA and protein relative expressions of toll like receptor-2( TLR2),nuclear factor κB( NF-κB) p65,myeloid differentiation factor 88( My D88) were compared. Results A total of 40 rats were modeled successfully. The levels of serum interleukin-6( IL-6),tumor necrosis factor-α( TNF-α),interleukin-1β( IL-1β) and the mRNA and protein relative expressions of TLR2,MyD88,NF-κB p65 were compared between each 2 groups,and the results showed that the indexes of the control group were the lowest,the middle dose group the second,the low and high dose groups slightly higher,the model group highest. There were significant differences between each 2 groups( P〈0. 05),without differences between the low and high dose groups( P〉0. 05). The levels of serum interleukin-6( IL-6),tumor necrosis factor-α( TNF-α),interleukin-1β( IL-1β) were compared among groups,and the 3 dose groups showed a significant decrease with time( P〈0. 05),while the model group showed a significant increase with time( P〈0. 05),and there were no significant differences in the control group with time( P〉0. 05). Conclusion Inula Cappa aqueous extract can improve the inflammatory response of SP rats,and the improvement effect of 12 g/kg of Inula Cappa aqueous extract is best,which may be related to the inhibition of TLR2/MyD88/NF-κB signaling pathway.
