摘要
目的采用HPLC法同时检测脂溶性红色素(苏丹红Ⅰ、苏丹红Ⅱ、苏丹红Ⅲ、苏丹红Ⅳ、颜料橙5)的含量,并用于检测中药及饮片非法添加的色素。方法选用资生堂CapcellPAKC18 ACR色谱柱(250mm×4.6mm,5μm),以乙腈-水为流动相进行梯度洗脱,流速1.0mL·min^-1,柱温为30℃,在不同波长下(480、508nm)进行检测。结果1~50μg·mL^-15种脂溶性红色素的线性关系良好,精密度、稳定性的RSD均小于5%,加样回收率为80%~120%。结论所用方法操作简单、快速、准确、重复性好、灵敏度高,可用于筛查中药材及饮片中脂溶性红色素的非法染色。
OBJECTIVE To establish a method of HPLC for simultaneously detecting fat - soluble red pigments ( sudan Ⅰ , sudan Ⅱ , sudan Ⅲ, sudan Ⅳ, pigment orange 5 ) , and to detect illegally added pigments in Chinese medicinal materials and pieces. METH- ODS The Shiseido CAPCELL PAK C18 column (250 mm × 4.6 ram,5 μm)was used to separate the samples at 30 ℃ with a step linear gradient using acetonitrile -water at a flow rate of 1 mL·min^-1 ,and detection was performed at different wavelengths (480,508 nm). RESULTS Five kinds of fat - soluble red pigments had good linear relationship in the range of 1 - 50 μg· mL^- 1, the RSD of precision and stability were less than 5% ,and the recovery was between 80% and 120%. CONCLUSION The method is simple, rapid and accurate with good reproducibility and high sensitivity, and it is suitable for the screening of the illegal dyeing of fat - soluble red pigments in traditional Chinese medicine and herbs.
