HPLC法同时测定排石颗粒中3个酚酸类成分的含量

Simultaneous determination of three phenolic acids in Paishi granules by HPLC

摘要 目的:建立同时测定排石颗粒中咖啡酰甘醇酸、迷迭香酸和丹酚酸A 3个酚酸类成分含量的高效液相色谱法。方法:采用Sun Fire C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈(A)-0.1%甲酸水溶液(B)为流动相,梯度洗脱,流速1.0 m L·min^(-1),检测波长330 nm,柱温30℃。结果:咖啡酰甘醇酸、迷迭香酸和丹酚酸A进样量分别在0.026 8~0.535 0μg(r=0.999 9)、0.261 8~5.236 0μg(r=0.999 9)和0.010 2~0.203 6μg(r=0.999 9)范围内与峰面积呈现良好的线性关系;平均回收率(n=6)分别为95.6%、99.0%和97.6%,RSD分别为1.1%、1.1%和1.4%;13批样品中咖啡酰甘醇酸、迷迭香酸和丹酚酸A含量范围分别为15.06~25.33、118.24~264.13和8.78~18.30μg·g^(-1)。结论:该方法可用于排石颗粒的质量控制。 Objective:To establish an HPLC method for the simultaneous determination of three phenolic acids in Paishi granules,namely,caffeoylglycolic acid,rosmarinic acid and salvianolic acid A.Methods:The Sun Fire C_(18) column(250 mm×4.6 mm,5μm)was used,the mobile phase was acetonitrile(A)-0.1%phosphoric acid(B)with gradient elution at a flow rate of 1.0 m L·min^(-1).The detection wavelength was 330 nm and the column temperature was at 30℃.Results:The linear ranges of caffeoylglycolic acid,rosmarinic acid and salvianolic acid A were 0.026 8-0.535 0μg(r=0.999 9),0.261 8-5.236 0μg(r=0.999 9)and 0.010 2-0.203 6μg(r=0.999 9),respectively.The average recoveries(n=6)were 95.6%(RSD=1.1%),99.0%(RSD=1.1%)and 97.6%(RSD=1.4%)respectively.The contents of caffeoylglycolic acid,rosmarinic acid and salvianolic acid A in thirteen batches of samples were 15.06-25.33μg·g^(-1),118.24-264.13μg·g^(-1) and 8.78-18.30μg·g^(-1),respectively.Conclusion:The method can be used to control the quality of Paishi granules.
