摘要
为了解棘腹蛙TGF-α基因信息和组织表达特性,采用RT-PCR克隆棘腹蛙TGF-α全长序列,发现棘腹蛙TGF-αc DNA全长1 021 bp,开放阅读框为486 bp,编码161个氨基酸,预测分子质量为17.28 k Da,其氨基酸序列与其他物种的相似性为65%~73%;利用实时荧光定量PCR方法对该基因的表达情况进行分析,发现在肝脏和胃脏有较高表达。本研究克隆了棘腹蛙TGF-α并对其组织分布进行了分析,为后期深入挖掘棘腹蛙TGF-α的生物学功能提供了参考。
For better understanding of the TGF-α encoding gene and tissue distribution characteristics,we studied the full length sequence of TGF-α from Paa boulengeri by RT-PCR. the results showed that TGF-α-Pb was 486 bp,encoding α 161 aa protein.The predicted molecular mass was 17. 28 k Da,and the amino acid sequence similarity with other species was 64-73%. Tissue distribution analysis of this gene by real-time fluorescent quantitative PCR method showed that it had higher expression in the liver and stomach. In this study,TGF-α-Pb from Paa boulengeri was cloned and the tissue distribution was analyzed.
