姬松茸多糖诱导巨噬细胞释放NO的机制 被引量:5

摘要 以小鼠巨噬细胞RAW264.7为研究对象,研究姬松茸多糖对其NO释放和iNOS表达的作用,并通过检测IκBα蛋白磷酸化水平的变化来探究姬松茸多糖诱导巨噬细胞释放NO的分子机制。结果表明,姬松茸多糖可剂量依赖性增强RAW264.7细胞NO的释放与iNOS蛋白的表达,且两者趋势一致。此外,25μg/mL姬松茸多糖能够增强RAW264.7细胞中p-IκBα蛋白的表达量,且在45min时达到最高,证明其能够激活NF-κB信号转导通路。综上,姬松茸多糖通过NF-κB途径上调巨噬细胞iNOS的表达,进而促进NO释放。 In this study,macrophage RAW264.7 cells were used to study the effect of Agaricus brasiliensis polysaccharide on NO release and iNOS expression,and the phosphorylation level of IκBαprotein was detected to explore its mechanisms.All the studies in the present paper could provide a new theoretical basis for the development of A.brasiliensis polysaccharide.The results showed that different concentrations of the polysaccharides enhanced the NO release and the expression of iNOS protein in RAW264.7 cells in the same dose-dependent manner.In addition,A.brasiliensis polysaccharide at 25μg/mL promoted the expression of p-IκBαprotein in RAW264.7 cells,and the highest expression was in 45 min(0 ~ 60 min),demonstrating that it could activate NF-κB signal transduction pathway.In conclusion,A.brasiliensis polysaccharides enhanced the release of NO in macrophages through up-regulating the iNOS expression by NF-κB pathway.
