HPLC法测定特殊医学用途婴配食品中视黄醇棕榈酸酯和视黄醇醋酸酯 被引量:6

摘要 采用正相高效液相色谱法测定特殊医学用途婴配食品中视黄醇棕榈酸酯和视黄醇醋酸酯的含量。采用氨基柱(150mm×4.6mm,5.0μm)为色谱柱,流动相:正己烷-25%叔丁基甲醚正己烷(体积比955),等梯度洗脱;流速:1.0mL/min,柱温:30℃;进样量:50μL;紫外检测器波长:325nm。结果表明:视黄醇棕榈酸酯保留时间为3.5min,视黄醇醋酸酯保留时间为6.0min,在0.05~10.0μg/mL呈线性相关(R2=0.999 9),检出限分别为2.0,2.5μg/100g。特殊医学用途婴儿配方食品中视黄醇棕榈酸酯含量为0.0~817.0μg/100g,精密度为0.9%~9.4%,视黄醇醋酸酯含量为0.0~8 683.0μg/100g,精密度为3.3%~6.3%,不同添加浓度回收率为81.0%~90.0%。该方法简单、快速、准确、精密度高,分离度好。 Retinol palmitate and retinol acetate in formulas for special medical purposes intended for infants was determined using NPHPLC.The HPLC system consisted of NH2 column(150 mm×4.6 mm,5.0μm),mobile phase of n-hexane-25.0% tert-butyl methyl ether n-hexane solution(95:5),flow rate of 1.0 mL/min,injection volume of 50 μL,column temperature of 30 ℃,UV detector detection wavelength of 325 nm.Results showed that retention time of retinol palmitate and retinol acetate were 3.5 min and6.0 min,respecitively the linearity was good in the range of 0.05~10.0μg/mL with a correlation coefficient of 0.999 9,the detection limits were 2.0 and 2.5μg/100 g respectively.The content of retinol palmitate and retinol acetate in formulas for special medical purposes intended for infants were 0~817.0 and 0~8 683.0μg/100 g,the precision were 0.9%~9.4% and 3.3%~6.3% respecitively.The recoveries of spiked samples at three levels were 81.0%~90.0%.The developed method presented the benefits of simplicity,rapidity,good separation,high accuracy,high precision.
