子宫肉瘤组织中SULT1E1与雌激素受体蛋白的表达及其相关性 被引量:2

摘要 目的探讨子宫肉瘤组织中SULT1E1与雌激素受体蛋白(ERα和ERβ)的表达水平及其相关性。方法选择62例明确诊断子宫肉瘤患者组织为观察组,根据其病理类型,分为子宫内膜间质肉瘤(ESS)和子宫平滑肌肉瘤(UL),其中ESS 43例,UL 19例;另选取正常子宫肌层组织20例为对照组。分别采用实时定量荧光PCR和免疫蛋白印迹技术检测各组患者组织中SULT1E1、ERα和ERβmRNA蛋白相对表达水平,进一步了解各蛋白表达水平在子宫肉瘤中的相关性。结果观察组患者组织中SULT1E1 mRNA蛋白较对照组显著降低(P<0.05),ERαmRNA蛋白较对照组显著降低(P<0.05),ERβmRNA蛋白表达水平与照组患者间未见明显差异(P>0.05)。而观察组患者ESS与UL亚型SULT1E1、ERα和ERβmRNA蛋白表达水平间未见明显差异(P>0.05)。在ESS组织中,SULT1E1与ERα呈负相关;在UL组织中,SULT1E1、ERα、ERβ之间均无线性相关性。结论子宫肉瘤组织SULT1E1蛋白表达降低或缺失可能参与了子宫肉瘤的发生与发展;在子宫内膜间质肉瘤组织中SULT1E1的表达与ERα表达呈负相关,提示它们的异常表达可能与子宫内膜间质肉瘤的发生、发展有关。 Objective To detect the expression and correlation of SULT1 E1 and estrogen receptor protein in uterine sarcoma.Methods A total of 62 cases of diagnosed uterine sarcoma selected as the observation group.According to their pathological types,they were divided into endometrial stromal sarcoma(ESS)and leiomyosarcoma of uterus(UL),including ESS in 43 cases and UL in 19 cases.In addition,20 cases of normal uterine myometrium were selected as the control group.The relative expression levels of SULT1 E1,ERαand ERβmRNA protein in each group were detected by real-time quantitative fluorescence PCR(qRT-PCR)and Western blotting.Meanwhile,the correlation between the expression levels of various proteins in uterine sarcoma was further understood.Results SULT1 E1 mRNA protein in the observation group was significantly lower than that in the control group(P〈0.05),and the ERαmRNA protein was significantly lower than that in the control group(P〈0.05),and the expression level of ERβmRNA there were not statistical significantly differences from that in the control group(P〈0.05).In the observation group,there were no statistical significantly differences between the ESS and the UL subtypes of SULT1 E1,ERαand ERβmRNA protein in the observation group(P〉0.05).In ESS tissue,SULT1 E1 was negatively correlated with ERα,and there was no linear correlation between SULT1 E1,ERαand ERβin UL tissues.Conclusion The expression of SULT1 E1 protein in uterine sarcoma tissue decreased or absent may be involved in the occurrence and development of uterine sarcoma.In the endometrium between SULT1 E1 sarcoma tissues and ERαexpression was negatively correlated,and suggested that abnormal expression may be related to endometrial stromal sarcoma.
