摘要
目的探究恩度联合紫杉醇对C57BL/6肺癌小鼠肿瘤生长及CAⅨ、HIF-1α表达的影响。方法将300只C57BL/6肺癌小鼠分成5组,分别为空白对照组、正常对照组、紫杉醇低剂量组、紫杉醇中剂量组和紫杉醇高剂量组,正常对照组注射7.5 mg/m^2恩度进行治疗,空白对照组注射等量生理盐水,紫杉醇低剂量组、紫杉醇中剂量组和紫杉醇高剂量组在正常对照组的基础上分别注射30、100、170 mg/m^2的紫杉醇,测量小鼠肿瘤的生长情况,测定小鼠的瘤重和抑瘤率,检测各组小鼠肺癌组织以及血清中CAⅨ、HIF-1α的表达水平。结果紫杉醇组的肿瘤体积显著小于空白对照组和正常对照组(P<0.05),且随着紫杉醇剂量的增加,肿瘤体积的减小程度明显增加(P<0.05);持续治疗4周后,紫杉醇组的瘤重显著小于对照组,抑瘤率显著高于对照组(P<0.05),且随着紫杉醇剂量的增加,瘤重的减小程度及抑瘤率的升高程度明显增加(P<0.05);紫杉醇组小鼠CAⅨ和HIF-1α的表达水平显著低于对照组(P<0.05);且随着紫杉醇剂量增加,CAⅨ和HIF-1α的表达水平显著降低(P<0.05)。结论恩度联合紫杉醇治疗C57BL/6小鼠肺癌能显著调控肿瘤的生长,降低CAⅨ和HIF-1α的表达水平,为肺癌的临床治疗提供一定的参考依据。
Objective To explore the influence of endostar combined with paclitaxel on tumor growth and expression of CA Ⅸ and HIF-1α in C57 BL/6 mice with lung cancer. Methods Totally 300 C57 BL/6 lung cancer mice were divided into five groups: blank control group,normal control group,paclitaxel low-dose group,paclitaxel mediumdose group and paclitaxel high-dose group. Normal control group were injected with 7. 5 mg/m^2 of endostar,blank control group were injected with the same volume of normal saline, and paclitaxel low-dose group,paclitaxel medium-dose group and paclitaxel high-dose group were injected with 30 mg/m^2, 100 mg/m^2 and 170 mg/m^2 paclitaxel respectively on the basis of normal control group. The growth of tumor in mice was measured, tumor weight and tumor inhibitory rate in mice were tested,and CA Ⅸ and HIF-1α expression levels of C57 BL/6 in lung cancer tissues and serum of mice were detected. Results Tumor volume of paclitaxel groups was significantly smaller than those of blank control group and normal control group( P 〈0. 05), and with the increase of paclitaxel dose, the tumor volume decreased significantly( P〈0. 05). After 4 weeks of continuous treatment, tumor weight in paclitaxel group was significantly less than that in control groups, and the inhibition rate was significantly higher than that in control groups( P〈0. 05), and with the paclitaxel dose increasing, the decreasing degree of tumor weight and increasing degree of tumor inhibition rate significantly increased( P〈0. 05). The expression levels of CA Ⅸ and HIF-1α in paclitaxel groups were significantly lower than those in control group( P〈0. 05), and with the paclitaxel dose increasing, the expression levels of CA and HIF-1 significantly decreased( P〈0. 05). Conclusion Endostar combined with paclitaxel can significantly regulate the growth of tumor in the treatment of C57 BL/6 lung cancer mice, and reduce CA Ⅸ and HIF-1 expression levels. It can provide some reference for clinical treatment of lung cancer.
