茜草炭复合提取物对子宫内膜异位症模型大鼠在位内膜增殖、凋亡及内膜组织VEGF、NF-κB的影响 被引量:6

摘要 目的探讨茜草炭复合提取物(QCS)对子宫内膜异位症模型大鼠在位内膜增殖、凋亡及内膜组织中血管内皮生长因子(VEGF)、核转录因子-κB(NF-κB)表达的影响。方法采用自体移植法建立子宫内膜异位症大鼠模型;将造模成功的大鼠随机分为假手术组、模型组、孕三烯酮组、QCS低剂量组、QCS中剂量组、QCS高剂量组。干预4周后,观察并记录各组异位病灶体积变化;免疫组化法检测增殖细胞核抗原(PCNA),TUNEL技术检测在位内膜细胞的增殖、凋亡;免疫组化法检测VEGF及NF-κB蛋白的表达,逆转录聚合酶链反应(RTPCR)技术检测NF-κB mRNA的表达。结果治疗后病灶体积减小,孕三烯酮组及QCS中、高剂量组均较模型组显著降低(P<0.05),且高剂量组效果显著优于孕三烯酮组(P<0.05);孕三烯酮及QCS中、高剂量均能显著促进在位内膜细胞凋亡,抑制增殖(P<0.05),且高剂量组效果显著优于孕三烯酮组(P<0.05);孕三烯酮组及QCS中、高剂量组在位内膜VEGF、NF-κB蛋白及mRNA的表达显著下调(P<0.05),且高剂量组下调效果显著优于孕三烯酮组(P<0.05)。结论茜草炭复合提取物能通过促进在位内膜凋亡、抑制其增殖及下调内膜组织中VEGF、NF-κB蛋白及mRNA的表达等机制治疗子宫内膜异位症。 Objective To explore the influence of madder and charcoal mixed extract( QCS) on eutopic endometrium proliferation and apoptosis and the expression of intimal tissue vascular endothelial growth factor( VEGF)and nuclear factor-κB( NF-κB) in model rats with endometriosis. Methods Endometriosis rats model was built by autotransplantation,and the rats modeled successfully were divided into sham operation group,model group,gestrinone group,QCS low-dose group,QCS medium-dose group and QCS high-dose group randomly. After 4 weeks' intervention,v olume change of ectopic foci in each group was observed and recorded. Proliferating cell nuclear antigen( PCNA)was detected by immunohistochemical method, and eutopic endometrial cells proliferation and apoptosis was tested by TUNEL technology. VEGF and NF-κB protein expression was detected by immunohistochemical method. NF-κB expression was tested by reverse transcription polymerase chain reaction( RT-PCR) technique. Results The volume of lesions decreased after treatment, and the lesion in gestrinone group,QCS medium-dose group and QCS high-dose group decreased significantly compared with that in model group( P〈0. 05), and QCS high-dose group was better than that of gestrinone group( P〈0. 05). Gestrinone and medium-dose and high-dose of QCS all significantly promoted the apoptosis and inhibited the proliferation in eutopic endometrium( P〈0. 05), and the effect in QCS high-dose group was better than that in gestrinone group( P〈0. 05). Gestrinone and medium-dose and high-dose of QCS significantly down-regulated the expression of protein and mRNA in VEGF and NF-κB in eutopic endometrium, and the down-regulation effect in QCS high-dose group was better than those in gestrinone group( P〈0. 05). Conclusion Madder and charcoal mixed extract can promote eutopic endometrial apoptosis, inhibit the proliferation, and down-regulate the expression of protein and mRNA in VEGF and NF-κB in endometrial tissue,which is the mechanism of the treatment of endometriosis.
