微小RNA-29b在非小细胞肺癌中的表达及其抑制A549细胞增殖的作用 被引量:1

Expression of microRNA-29b in non-small cell lung cancer and its function of inhibiting A549 proliferation

摘要 目的研究miR-29b在非小细胞肺癌(NSCLC)组织中的表达及其在肺腺癌细胞A549细胞株中的生物学功能。方法选择2013年6月至2015年5月河南大学淮河医院手术切除的NSCLC患者40例为研究对象,用实时荧光定量PCR聚合酶链反应法检测40例NSCLC患者癌组织和癌旁组织石蜡标本中的miR-29b的表达水平并探讨其与患者临床病理学特征的关系。用脂质体法将外源性miR-29b模拟物miR-29b mimics转染至人肺腺癌细胞株A549,用CCK-8实验和Transwell实验评价miR-29b mimics对A549细胞增殖及侵袭能力的影响。结果 40例NSCLC患者癌组织和癌旁组织中miR-29b的表达水平分别为-1.74±1.26,-0.63±0.54,癌组织显著低于癌旁组织,差异有统计学意义(P<0.05)。miR-29b表达水平与患者的年龄、性别、病理类型、分化程度和区域淋巴结转移无关(P>0.05),但与患者的临床分期有关(P<0.05);Ⅰ/Ⅱ期患者miR-29b表达水平显著高于Ⅲ期,差异有统计学意义(P<0.05)。与转染前相比,转染外源性miR-29b后A549细胞中miR-29b表达水平显著增高;转染miR-29b mimics后,A549细胞增殖能力显著降低,差异均有统计学意义(均P<0.05);但穿模能力无明显变化,差异无统计学意义(P>0.05)。结论微小RNA miR-29b在非小细胞肺癌患者癌组织中表达水平显著降低,过表达miR-29b可抑制肺癌A549细胞增殖。 Objective To observe miR-29 b expression in lung cancer tissue and normal lung tissue of non-small cell lung cancer( NSCLC)patients and its association with patients clinical pathological features,discussing miR-29 b biological function in A549 cells. Methods Forty NSCLC patients were included in this study from Jun 2013 to May 2015.MiR-29 b expression level were assayed by real-time PCR for cancer and normal lung tissue. MiR-29 b expression and patients ' clinical pathological features were analyzed. MiR-29 b mimics were transfected into A549 cell. After transfection,the cell proliferation and migration ability were tested by CCK-8 and transwell assay. Results MiR-29 b expression level were-1. 74 ± 1. 26,-0. 63 ± 0. 54 for cancer and normal lung tissue with statistical difference( P〈0. 05); miR-29 b expression was not associated with patients' age,gender,pathology type and lymph node metastasis( P〉0. 05). However,the expression level of miR-29 bin stage Ⅰ/Ⅱ patients was significantly higher than that in stage Ⅲ patients( P〈0. 05). MiR-29 b was overexpressed by transfected miR-29 b mimics in A549 cells. After transfection,the A549 cell proliferation ability was significant inhibited( P〈0. 05). However,its migration ability was not changed( P〉0. 05). Conclusion MiR-29 b was low expressed in lung cancer tissue and overexpression miR-29 b can inhibit A549 cell proliferation.
