几丁糖对鼠坐骨神经损伤后再生的影响 被引量:2

Effects of chitosan on regeneration of rat with sciatic nerve injury

摘要 目的研究局部用几丁糖对促进大鼠坐骨神经再生的影响。方法按照体重将大鼠随机分为2组:模型组(12只)和实验组(12只)。以离断大鼠左侧坐骨神经建立模型。建模成功后,模型组大鼠坐骨神经吻合口周围滴注0.9%NaCl 50μL;实验组大鼠坐骨神经吻合口周围滴注液态几丁糖50μL,使几丁糖包绕坐骨神经吻合口。用免疫印迹法检测坐骨神经吻合口的Ⅰ型、Ⅲ型胶原蛋白含量,用苏木精-伊红(HE)染色观察坐骨神经再生情况。结果给药后4周,实验组和模型组的Ⅰ型胶原蛋白含量分别是0.22±0.06,0.28±0.03;这2组的Ⅲ型胶原蛋白的含量分别是0.47±0.06,0.36±0.06,实验组与模型组比较,差异均有统计学意义(P<0.01,P<0.001)。这2组的再生神经纤维数量分别是(80.33±7.37),(54.33±12.06)n,实验组与模型组比较,差异有统计学意义(P<0.05)。结论医用几丁糖凝胶可预防神经周围黏连,具有促进周围神经再生的作用。 Objective To study of the effect of chitosan on the promote regeneration of sciatic nerve in rats. Methods The Sprague-Dawley( SD) rats were randomly divided into 2 groups: model group and experimental group. The rats in both groups were severed the left sciatic nerve to establish model. Model group,dripped with 50 μL normal saline after the successful modeling,and the experimental group was injected with 50μL liquid chitosan around the anastomosis mouth after the model was successfully established. The content of type Ⅰ and type Ⅲ collagen in sciatic nerve anastomosis were detected by Western blot. The sciatic nerve regeneration was observed by hematoxylin eosin( HE) staining.Results After 4 weeks of administration,the contents of type Ⅰ in experimental group and model group were 0. 22 ± 0. 06,0. 28 ± 0. 03; the type Ⅲ collagen in the 2 groups were 0. 47 ± 0. 06,0. 36 ± 0. 06; comparison between experimental group and model group,the difference was significant( P〈0. 01,P〈0. 001). The number of regenerated nerve fibers counted by sciatic nerve in the 2 groups were( 80. 33 ± 7. 37),( 54. 33 ± 12. 06) n; comparison between experimental group and model group,the difference had significantly( P〈0. 05). Conclusion Medical chitosan gel can prevent perineural adhesions and promote peripheral nerve regeneration.
