摘要
目的:对祁山药中的多酚类物质进行提取,确定最佳提取工艺(料液比、提取温度、提取时间、丙酮体积分数),并对提取液中的祁山药多酚用Folin_Ciocalteu(福林酚)试剂比色法进行含量测定。方法:通过单因素试验法(料液比、丙酮的体积分数、提取时间和温度的变化)来确定提取祁山药中多酚时每个因素的最佳提取条件,再用正交试验法来确定提取山药中多酚时的提取条件的最优组合。结果:祁山药多酚的最优提取工艺:提取溶剂用体积分数为50%的丙酮,料液比1∶20,提取时间80min,提取温度55℃。结论:试验方法简便、操作简单、准确度高,能够用来测定祁山药以及其他所含多酚类物质的植物中的多酚含量,并且还能为山药多酚的深入研究和综合开发利用提供理论依据。
Objective: In order to extract the polyphenols in Chinese Dioscorea opposita Thunb, and determine the optimal extraction process (liquid ratio, extraction temperature, extraction time, and acetone volume fraction). The content of polyphenols in the extracted liquid was measured by Folin_Ciocalteu (forinol) reagent colorimetry. Method: The optimal extraction conditions of polyphenols from Chinese Dioscorea opposita Thunb were determined by single factor test (ratio of liquid to material, volume fraction of acetone, extraction time and temperature change). Orthogonal test was used to determine the optimal combination of extraction conditions for polyphenol extraction from Chinese Dioscorea opposita Thunb. Result : The optimum extraction process of polyphenols from Chinese Dioscorea opposita Thunb was detemlined by this experiment. The liquid to liquid ratio is 1 : 20. The extraction time is 80 minutes; Extraction temperature was 55℃. Conclusion: The test method is simple, the operation is simple, high accuracy, can be used to determination of yam and other polyphenols contained polyphenols content in plants, and also for the further research of Chinese Dioscorea opposita Thunb polyphenols and provide theoretical basis for comprehensive development and utilization.
