长链非编码RNA-HULC调控肺腺癌细胞的增殖和侵袭 被引量:1

摘要 目的:探讨长链非编码RNA-HULC(long non-coding RNA-HULC,lnc RNA-HULC)对肺腺癌细胞增殖和侵袭过程的调控作用。方法:收集57例肺癌患者的癌组织及与其配对的癌旁组织,通过实时定量PCR(quantitative real-time PCR,q RT-PCR)法检测组织及细胞中lnc RNA-HULC的表达水平;采用RNA干扰技术(si RNA)下调lnc RNA-HULC的表达,同时以q RT-PCR法检测其干扰效率。CCK-8法和Transwell法检测下调lnc RNA-HULC对肺腺癌细胞增殖和侵袭过程的影响。结果:与癌旁组织相比,lnc RNA-HULC在癌组织中的表达显著升高;与正常肺上皮细胞相比,lnc RNA-HULC在肺癌细胞中的表达也显著升高。下调lnc RNA-HULC的表达后,肺腺癌细胞的增殖和侵袭能力均降低。结论:下调lnc RNA-HULC可抑制肺腺癌细胞的增殖和侵袭过程。 Objective: To investigate the potential molecular mechanism of long non-coding RNA-HULC(lnc RNAHULC) in the proliferation and invasion of lung adenocarcinoma cell lines. Methods: Quantitative RT-PCR was used to detect the expression of lnc RNA-HULC in lung cancer tissues and cell lines. Small interfering RNA(si RNA) was used to down-regulate lnc RNA-HULC expression. CCK-8 assay and Transwell assay were performed to explore cell proliferation and invasion. Results: Compared with adjacent tissues, the expression of lnc RNA-HULC was increased in lung cancer tissues. Compared with normal lung epithelial cell, lnc RNAHULC was also increased in lung cancer cell lines. Silence of lnc RNA-HULC inhibited cell lines proliferative and invasive ability. Conclusion: Down-regulation of lnc RNA-HULC can cell proliferation and invasion in suppress lung adenocarcinoma.
