虾青素-胶原蛋白耦合物改善小鼠皮肤光老化作用研究 被引量:3

Studies on improvement of astaxanthin-collagen peptide coupling on mice skin photoaging damage

摘要 目的在制备虾青素-胶原蛋白耦合物的基础上,探讨耦合物对小鼠皮肤光老化的改善作用。方法将来源于雨生红球藻的虾青素单体和均一分子量胶原蛋白多肽在一定条件下反应后生成耦合物,将耦合物与适当基质混合后制成外用乳膏。采用UV灯照射Balb/c小鼠背部裸露皮肤,建立动物光老化模型,随机分为正常对照组、模型对照组、基质对照组、虾青素组、胶原蛋白多肽组、耦合物组和维生素E组,每天UV照射前进行药物涂抹。6周后观察小鼠皮肤形态、测定皮肤厚度变化及进行皮肤组织病理学检测等。结果虾青素和胶原蛋白可结合生成具有特征吸收波长的水溶性耦合物,耦合物可有效缓解皮肤厚度增生,修复皮肤胶原纤维和弹性纤维网状结构。结论虾青素-胶原蛋白多肽耦合物可有效改善UV引起的皮肤光老化损伤。 Objective To investigate the effect of coupling on skin photoaging based on the preraration of astaxanthin-collagen peptide coupling.Methods Astaxanthin separated from Haematococcuspluvialis and collagen peptide with homogenous molecular weight distribution were combined to form coupling,and the coupling was mixed with matrix to prepare an external cream.UV light was used to irradiate bare skin on the back of Balb/c mice to establish animal model of photoaging,drugs were applied to the mice on the back before UV irradiation daily.The Balb/c mice were randomly divided into seven groups including normal control group,model control group,ointment base control group,astaxanthin group,collagen peptide group,astaxanthin-collagen coupling group and vitamin E group.After 6 weeks,the mice skin on the back were observed for the general morphology,the measure of skin thickness and histopathological observation.Results Astaxanthin and collagen were combined to form water-soluble coupling with characteristic absorption,the coupling improved skin thickness hyperplasia and restored the network of collagen fiber and elastic fiber.Conclusion Astaxanthin-collagen peptide coupling could effectively improve UV-induced skin photoaging.
