5种北虫草子实体中核苷类有效成分的提取工艺优选及含量测定

摘要 目的优选北虫草子实体中3种核苷类成分提取工艺,建立其含量测定方法;并比较5种北虫草子实体中主要核苷类成分的差异。方法采用RP-HPLC法测定北虫草子实体中尿苷、腺嘌呤、虫草素的含量;在单因素试验基础上,采用L_9(3~4)正交试验考察甲醇体积分数、提取时间、提取温度及料液比对北虫草子实体中3种核苷类成分提取工艺的影响。结果最佳提取工艺为20倍量水于35℃下超声提取30min;5种北虫草子实体中主要核苷类成分含量有较大差异。结论优选的提取工艺重复性好、稳定可行。建立的含量测定方法可用于北虫草子实体中核苷类成分的定性定量研究,评价不同来源虫草的质量。 Objective To optimize the extraction technology of three nucleosides from codyceps militaris fruitbodies and establish the content determination method. To compare the contents of nueleosides from five codyceps militaris fruitbodies. Methods The contents of uridine, adenine and eordycepinwere determined by RP-HPLC. Effects of the volume fraction of methanol, extractiontime, extraction temperature, solid-liquid ratioon yield of three nucleosides were investigated by single factor tests, and then extraction technology was optimized by orthogonal test. Results Optimum ultrasonic extraction conditionswere as follows:codyceps militaris fruitbodies was extracted at 35℃ with 20 times water for 30 min. The contents of major nueleosides in 5 cordyceps militaris fruitbodies varied greatly. Conclusions The optimized extraction technology has good repeatability, stability and feasibility. The estab- lished method of content determination can be used forqualitative and quantitative study of nucleosides in cordyceps militaris fruitbodies, and evaluate the quality of cordyceps sinensis from different sources.
