骨髓间充质干细胞对视网膜脱离兔脑源性神经营养因子表达的影响

The effects of the bone marrow mesenehymal stem cells on the expression of brain derivedneurotrophic factor in rabbits with retinal detachment

摘要 目的观察骨髓间充质干细胞对视网膜脱离后视网膜组织中脑源性神经营养因子(BDNF)表达的影响。方法健康青紫蓝兔54只,随机分为3组:A组为正常对照组;B组为视网膜脱离(RD)+磷酸盐缓冲液(PBS)组;C组为RD+BMSC组,各组均为18只兔。建模后1、2、4周,分别各处死6只,眼球标本制作石蜡切片后用于HE染色及免疫组化染色。用图像分析软件Image J对视网膜组织切片的着色结果进行分析。结果HE染色切片显示:B组、C组建模后1周视网膜均复位,各时间段视网膜整体厚度及神经节细胞数目对比示:A组〉C组〉B组。免疫组化切片显示:3组各时间点均可见BDNF阳性表达,表达强度C组〉A组〉B组,差异有统计学意义(P〈0.05)。结论兔RD模型可在1周内解剖复位,但复位的视网膜细胞排列紊乱,兔RD模型视网膜组织中BDNF的表达可能对视网膜脱离后的视功能恢复有意义。 Objective To observe the effects of the bone marrow mesenehymal stem cells (BMSC) on the expression of brain derived neurotrophic factor (BDNF) in the retinal detachment (RD) of rabbits. Methods Fifty-four healthy rabbits were randomly divided into 3 groups,18 rabbits in each group. Group A were nomal control group, group B received retinal artifical hole and injection of phosphate buffered saline (PBS) ; group C received retinal artifical hole and injection of PBS with BMSC. At 1,2 and 4 weeks after modeling,6 rabits were killed in each group. The specimen of their eyeballs was made of paraffin section and used for HE staining and immunohistochemical staining. The colouring result of retinal tissue section were analyzed with Image J. Results The reattachment of retina were achieved at 1 week after modeling in group B and group C with HE staining. The overall thickness of the retina and the number of retinal ganglion cell were the most in group A, and followed by group C, and group B at each time point. The results of immunohistoehemistry showed that the expression of BDNF were positive in three groups. The expression of BDNF were the strongest in group C and followed up, group A and group B. The difference were statistically significant among the three groups ( P 〈 0. 05 ). Conclusion The retinal of RD model can be anatomic reattachemnt in 1 week, but the arrangement of retinal cells are disordered. The expression of BDNF in the RD model mav have significance meaninz for the recovery of visual function.
