海峡两岸荒川库蠓亲缘地理学研究

PHYLOGEOGRAPHIC STUDIES ON CULICOIDES ARAKAWAI FROM BOTH SIDES OF TAIWAN STRAITS

摘要 以COⅠ、16S r DNA和ITS基因为标记,选用环斑库蠓Culicoides circumscriptus(N267)作为外群,对海峡两岸10个群体77个荒川库蠓标本的遗传距离、基因分化、系统发育和种群结构进行分析。基于COⅠ、16S r DNA基因数据集及ITS基因数据集的AMOVA分析均显示,大的遗传变异主要来自种群内的遗传变异,在线粒体数据集及核糖体数据集的比例分别为96.75%和91.44%。不同基因的分子数据显示,不同种群间的荒川库蠓遗传分化程度低、基因交流频繁,各个地理种群没有按照地理区划形成独特的遗传结构。气流及人为因素可能是造成其远距离扩散的主要原因。 The mitochondrial gene (CO I, 16S rDNA) and nuclear gene (ITS1-ITS2) were employed as genetic markers to examine the genetic diversity and population structure of Culicoides arakawai in both sides of the Taiwan Straits. Molecular variance (AMOVA) analysis based on the concatenated dataset indicated that the genetic variance was attributed to the variation within populations with the proportion of mitochondrial genes and nuclear gene 96.75% and 91.44% , respectively. Phylogenetic analysis based on different molecular dataset clustered into three well supported lineages. Amongthem, lineage I was formed with haplotype YN1 and GX9, lineage Ⅱ was formed with haplotype AH5 and AH7, other haplotypes formed lineage Ⅲ. it was suggested that there were no significant genetic differences among populations. Airflow and human activities might be the main reasons for the spread of C. arakawai.
