摘要
建立了紫外光诱导角质细胞损伤及凋亡模型,并初步研究了珍珠提取物抑制紫外光诱导人表皮角质细胞凋亡。利用紫外光诱导人表皮角质细胞凋亡,实验分为对照、模型、和紫外光加药3组。采用cck8、Hoechst33258染色等方法检测紫外光诱导细胞后细胞形态以及细胞凋亡的状况;酶化学法测定细胞谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量。与模型组相比,紫外光加药组角质细胞的增殖活性显著升高,GSH-Px、SOD水平显著升高,MDA含量显著降低。珍珠提取物可在一定程度上抑制紫外光介导的人角质细胞的损伤以及凋亡。
This study aims to establish the cell injury and apoptosis model of Ha Ca T cells induced by ultraviolet radiation( UV),and preliminary study proves that the pearl extract inhibits ultraviole tradiation induced Ha Ca T cells apoptosis. Using UV induced Ha Ca T cells apoptosis,experiment design is divided into 3 groups,control group,model group,and pearl extract group. The viability of Ha Ca T cells was measured by CCK8 chromatometry,then the morphological changes of cells were observed through Hoechst 33258 staining,zymochemistry method was performed to detect the activity of GSH-Px、SOD、MDA content. Compared with model group,GSH-Px and SOD activity were increased significantly,the content of MDA was decreased significantly in pearl extract group. Pearl extract can protect Ha Ca T cells from UV-induced injury and apoptosis.
