新仙灵骨葆胶囊对去卵巢骨质疏松大鼠的影响 被引量:5

摘要 目的观察新仙灵骨葆胶囊对去卵巢大鼠骨质疏松的治疗作用,并探讨其作用机制。方法以去卵巢术制备大鼠骨质疏松模型,随机分为假手术组,模型组,尼尔雌醇对照组(0.7 mg·kg^(-1)),仙灵骨葆胶囊组(350 mg·kg^(-1)),新仙灵骨葆胶囊低、中、高剂量组(53,105,210 mg·kg^(-1)),连续灌胃给药12周。末次给药后分析大鼠的腰椎骨、股骨、胫骨的骨密度;生化指标检测血钙(S-Ca)、血磷(S-P)、尿钙(U-Ca)、尿磷(UP)、血清碱性磷酸酶(ALP)的含量;ELISA法测定血清骨特异性碱性磷酸酶(BALP)、骨钙素(OCN)含量及尿液脱氧吡啶啉(DPD)含量;计算子宫指数,检测股骨生物力学性能。结果与假手术组比较,模型组大鼠的体质量和U-Ca、U-P、ALP、BALP、OCN、DPD含量明显升高,子宫指数和S-Ca、S-P含量显著降低,大鼠椎骨、股骨、胫骨的骨密度及股骨生物力学性能显著降低。与模型组比较,各药物组均能有效抑制大鼠体质量增加,升高S-Ca、S-P含量,降低U-Ca、U-P、ALP、BALP、OCN、DPD含量,增加大鼠椎骨、股骨、胫骨的骨密度,改善大鼠的股骨生物力学性能。结论新仙灵骨葆胶囊通过抑制骨吸收对去卵巢大鼠所致的骨质疏松症有良好的治疗效果,且该作用一定程度上优于仙灵骨葆胶囊。 Objective To explore the effect and mechanism of New Xianling Gubao capsule(NXGC) on ovariectomy induced osteoporosis in rats. Methods The rat model of osteoporosis was induced by ovariectomy. The rats were evenly randomized into 7 groups,namely Sham operation group,model group,nilestriol positive group(0.7 mg·kg^(-1)),Xianling Gubao capsule group(XGC,350 mg·kg^(-1)),and low-dose,middle-dose and high-dose NXGC groups(with the dosees of 53,105,210 mg·kg^(-1) respectively). The corresponding drugs were given by oral administration with 10 m L·kg^(-1) for 12 w,and then the body weight of the animals was recorded weekly during the experimental period. Bone mineral density(BMD) of the lumbar vertebra,femur and tibia were measured. Bone turnover markers(Serum alka-line phosphatase(ALP),bone-specific alka-line phosphatase(BALP),osteocalcin(OCN),deoxypyridinoline(DPD) and other parameters including serum calcium(S-Ca),serum phosphorus(SP),urine calcium(U-Ca),phosphorus(U-P) were measured by biochemical or ELISA methods. Uterine were removed from each rat and immediately weighed,and the femur was taken for biomechanical measurements. Results According to the results, compared with the model group, NXGC and XGC significantly prevented ovariectomyinduced bone loss and the body weight gain without affecting the weight of the uterus,and increased S-Ca,S-P levels, decreased level of bone turnover markers and U-Ca, U-P, ALP, BALP, OCN, DPD levels in ovariectomized rats, and improved the biomechanics. And the effects of NXGC were better than that of XGC.Conclusion The protective effect of NXGC is due to an increased bone formation with reduction of bone resorption in ovariectomized osteoporotic rats,and the effect is better than that of XGC.
