健妇胶囊对雌激素、孕激素和促红细胞生成素水平的影响

Effect of Jianfu Capsule on The Levels of Estrogen,Progestogen and Erythropoietin

目的通过细胞活性实验研究健妇胶囊补血养血和调经止带的功效。方法选择人源胚胎肾细胞HEK293T细胞系,构建检测促红细胞生成素(EPO)表达调控立体研究模型,在该细胞转染缺氧反应组件—荧光素酶(p HRE-luciferase)载体,研究健妇胶囊的补血养血功效;选择MCF-7人乳腺癌细胞,构建雌激素反应元—荧光素酶活性测定(p ERE-luciferase)模型,测试健妇胶囊对雌激素(Estrogen)反应水平的影响;另外选择lshikawa(人子宫内膜腺癌细胞),测试健妇胶囊对孕激素受体(Progesterone receptor)表达的影响。从三个方面的研究评估"健妇胶囊"的功效。结果健妇胶囊的水提取物对HEK293T细胞系,MCF-7人乳腺癌细胞和lshikawa人子宫内膜腺癌细胞没有毒性;对人源胚胎肾细胞HEK293T有促红细胞生成素(EPO)生成的作用,显示具有补血功效,其补血效用有剂量依赖性,量越大,补血功效越强;对雌激素有调控作用,当健妇胶囊的浓度为1 mg·mL^(-1)时,可增强孕酮受体的表达。结论健妇胶囊具有增强促红细胞生成素(EPO)生成功效,证明其具有补血功效;对雌激素反应元ERE的激活,证明健妇胶囊对雌激素的调控作用;另外健妇胶囊对孕酮受体表达作用,证明健妇胶囊具有临床治疗生殖激素相关障碍的效果。

Objective This study investigated the effect of Jianfu capsule on blood supplements and bloodmaintenance, the effect of regulating the menstruation and arrest leucorrhea through cell activity experiment.Methods HEK293 T cell line of human embryonic kidney cell was selected to construct a three-dimensional modelfor detecting EPO expression and transfection of p HRE-luciferase carrier in the cell to study the blood-tonic effect of Jianfu capsule. MCF-7 human breast adenocarcinoma cells were selected,and the ERE-luciferase assay model wereestablished to test the effect of Jianfu capsule on estrogen response level. In addition,lshikawa human endometrialadenocarcinoma cells were selected to test the effect of Jianfu capsule on progesterone receptor expression. Results The water extract of Jianfu capsule had no toxicity to HEK293 T cell line,MCF-7 human breast adenocarcinoma celland shikawa human endometrial adenocarcinoma cell. Treatment with water extract of Jianfu capsule on humanembryonic renal cell（HEK293 T） was shown to have the effect of blood supplement,which was dose-dependent：the greater the quantity,the stronger the effect. It can regulate estrogen and enhance the expression of progesteronereceptor when the concentration of the water extract is 1 mg·mL^（-1）. Conclusion The study of the effect of Jianfu capsule on the production of erythropoietin（EPO） proved its blood-tonic effect. Activation of ERE,an estrogenresponse element,proved the estrogen regulatory effect of Jianfu capsule. In addition,the effect of Jianfu capsule onprogesterone receptor expression was demonstrated to be effective in treating reproductive hormone-related disorders.
