电针对肥胖大鼠脂肪组织脂联素与瘦素表达的影响 被引量:4

Effects of electroacupuncture on adiponectin and leptin expressions in adipose tissue of Sprague-Dawley rats with experimental obesity

摘要 目的探讨电针胃经穴位干预肥胖SD大鼠对脂肪组织脂联素(ADPN)和瘦素(LP)表达的影响。方法健康雄性清洁级SD大鼠36只,随机分为正常饮食组和高脂饮食组各18只,后者高脂饮食饲喂8 w。造模前后测量体重身长。正常饮食组随机分为正常电针组和正常对照组各9只,高脂模型组随机分为肥胖电针组和肥胖对照组各9只。造模结束后连续电针30 d,断头处死SD大鼠,迅速分离腹腔脂肪组织置入-80℃冰箱。Western印迹法检测各组大鼠脂肪组织ADPN与LP蛋白表达情况。结果造模8 w后,高脂饮食组体重、体重增长值及Lee指数与正常饮食组相比差异有统计学意义(P<0.05),显示肥胖模型复制成功。肥胖电针组电针后脂肪组织的ADPN蛋白表达水平显著高于肥胖对照组(P<0.05),LP蛋白的表达水平显著低于肥胖对照组,差异均有统计学意义(P<0.05)。正常电针组电针后脂肪组织ADPN和LP表达与正常对照组比较差异无统计学意义。结论足三里和内庭配伍电针可以改善肥胖大鼠脂肪组织的蛋白代谢状态。 Objective To investigate the effect of electroacupuncture( EA) on Zusanli and Neiting acupoint on adiponectin( ADPN) and leptin( LP) expressions in adipose tissue of Sprague-Dawley( SD) rats with experimental obesity.Methods 36 clean grade SD rats were randomly divided into high lipid and control groups,18 rats in each group. All experimental obesity rats were fed with high fat diet for8 weeks. Body weight and body length were measured before and after modeling. Then either high lipid group or control group was randomly divided into 2 groups respectively,as blank control and blank EA groups,obesity control and obesity EA groups,9 rats in each group. Rats of blank EA and obesity EA groups were treated with electro-acupunctured for 30 days,while other groups were fixed at the same time once per day. The rats were sacrificed with decapitation,while their adipose tissues were immediately separated and stored in-80℃ refrigerator.ADPN and LP expressions in adipose tissue were detected by Western blot.Results The weight,weight gain and Lee's index in rats fed by high fat diet were higher than those of control group( P0.05). ADPN expression in adipose tissue of obesity EA group was increased compared with that of blank control group( P0.05),LP expression in adipose tissue of obesity EA group was decreased( P0.05). The expressions of ADPN and LP in adipose tissue of blank EA group were not significantly varied,compared with those of blank control group( P 0.05).Conclusions EA at Zusanli and Neiting acupoint could improve the protein metabolism state of adipose tissue in obese rats.
