摘要
以栅藻S.raciborskii WZKMT为原料,采取5种预处理方法进行微藻破壁和多糖抽提探究,研究发现以硫酸处理法效果最佳,其最优条件为20g/L底物浓度、140℃、40 min和4%硫酸(w/v)。此外,应用傅里叶变换红外光谱(FTIR)分析酸处理前后藻体细胞的结构变化。最优条件下,葡萄糖和木糖的收率(质量分数)分别为85.8%和99.7%,葡萄糖和木糖含量可分别高达18.55 g/L和11.17 g/L。
Cellular disruption and sugars extraction by five different pretreatment methods with Scenedesmus raciborskii WZKMT were studied.The results showed that the best pretreatment was with acid pretreatment by 4% H_2 SO_4(w/v),withconcentration of 20 g/L algae,at 140 ℃ for 40 min.In addition,the structure of algae by sulfuric acid treated was alsoanalyzed with Fourier Transform Infrared Spectroscopy(FTIR).Under these conditions,the yield of glucose and xylosewere 85.8% and 99.7%(w/w),respectively.And the maximum concentration of glucose and xylose can attain to 18.55 g/Land 11.17 g/L,respectively,at loading of 160 g/L algae.
