摘要
目的:探讨覆盆子乙酸乙酯部位几种单体对H_2O_2诱导PC12细胞损伤的保护作用,筛选出覆盆子乙酸乙酯部位中抗细胞损伤效果较好的单体。方法:通过过氧化氢(H_2O_2)诱导PC12建立细胞损伤模型,进行MTT细胞毒性、Cell Counting Kit-8(CCK8)细胞增殖、细胞凋亡、细胞乳酸脱氢酶(LDH)、丙二醛(MDA)、活性氧(Reactive oxygen species,ROS)检测。结果:100μmol/L H_2O_2损伤PC12细胞时,细胞存活数量适当,形态完整,为最佳造模浓度;结果显示,模型对照组与空白对照组相比各项指标显著改变,槲皮素和山奈酚组PC12细胞的活性相较于模型对照显著提高,LDA、MDA、ROS含量显著增高,LDH、MDA与ROS释放相对减少。结论:H_2O_2诱导PC12细胞损伤模型的最佳浓度为100μmol/L;槲皮素和山奈酚对H_2O_2诱导的PC12细胞损伤具有一定的保护作用。
Objective: The aim of this study was to evaluate the different components of raspberry extracted by ethyl acetatec on improving the cognition impairment and screen the effective components of raspberry extracted by ethyl acetate. Methods: cellular damaged of PC12 cells induced by H_2O_2 was used to evaluated the anti-damaged effect of drugs,The cytotoxicity of MTT,the proliferation CCK8,the content of malondialdehyde(MDA),the activity of lactate dehydrogenase(LDH),the activitity of Reactive oxygen species(ROS) was detected. Results: The number and phenotype of PC12 treated with the concentration of 100μmol/L H_2O_2 were advisable. The MTT revealed that the activity of PC12 treated with quercetin and campherolwere increased significantly. The LDH demonstrated that after stimulating with H_2O_2,the impairment of PC12 treated with quercetin and campherol was decreased significantly and the concentration of LDH decreased,the MDA and ROS express the similar results. Conclusion: The optimum concentratioof H_2O_2 which was harm to PC12 was 100μmol/L. The quercetin and campherol serve a protective function to PC12 induced by H_2O_2.
