红芪不同提取物对局灶性脑缺血大鼠脑组织损伤的保护作用研究 被引量:2

摘要 目的:探讨红芪不同提取物对局灶性脑缺血大鼠的影响。方法:SPF级Wistar大鼠分为模型组、假手术组、银杏叶片组(0.21g/kg)、红芪醇提物组、水提物组、醇沉物组、残留物组,各提取物组灌胃量均为6.0g/kg,模型组和假手术组灌胃等量生理盐水。采用50%FeCl_3作用大脑中动脉造局灶性脑缺血大鼠模型;盲法进行神经功能评分;称重法检测脑含水量;TTC法检测脑梗死面积比;试剂盒检测脑组织中SOD、CAT、MDA及血清中NO、NOS、ET、CGRP含量;UV法测定多糖和总皂苷含量。结果:与模型组比较,红芪醇提物组、水提物组、醇沉物组脑含水量均显著降低;醇沉物组SOD、CAT显著升高,MDA显著下降;水提物组SOD显著升高;其他各指标组间无统计学意义。但直观分析,水提物组、醇沉物组神经功能缺损均有改善趋势;醇沉物组脑梗死面积比模型组有所降低;红芪水提物多糖(22.6%)和皂苷(7.7%)含量均较高,醇沉物多糖(42.2%)含量最高,皂苷(1.9%)。结论:6.0g/kg剂量的红芪水提物和醇沉物有降低急性局灶性脑缺血造成的脑组织及神经功能损伤的作用。这种作用可能是通过增强大鼠脑组织抗氧化功能、缓解血流异常变化、调节机体免疫功能等实现的,且总多糖和总皂苷应该是其主要活性物质基础。 Objective: To study the effect of different extracts of Radix Hedysari on neurological function. Methods: Wistar rats(SPF degree) were randomly divided into the model,the sham,ginkgo leaf dispersible tablet(0. 21 g/kg),ethanol extract,water extract,alcohol precipitation,residuum groups. Ten rats were in each group and rats were gavaged by corresponding extracts with 6. 0 g/kg. Rats with focal cerebral cerebral ischemia were established by 50% FeCl_3 in middle cerebral artery. Neurological function was scored by Bederson and Longa. Brain water content was detected by weight method. Brain infarction area was detected by TTC staining method. SOD,CAT,MDA in brain tissues and serum NO,NOS,ET,CGRP were detected by kits. Polysaccharide and total saponins were determinated by ultraviolet spectrophotometry.Results: Compared with the model group,brain water contents in ethanol extract,water extract and alcohol precipitation groups were reduced(P〈0. 01). SOD and CAT in alcohol precipitation group were rised(P〈0. 05). MDA in alcohol precipitation group was dramatically declined(P〈0. 05). SOD in water extract group was rised(P〈0. 05). Compared with the model group,there were no statistically significant differences in other indicators in other groups. But through intuitive analysis,we found that water extract and alcohol precipitation had slight efftct on improving neurologic impairment. And brain infarction area in alcohol precipitation group was lower than that in the model group.Water extract and alcohol precipitation slightly improved contents of SOD,CAT,MDA,NO,NOS,ET,CGRP. Contents of polysaccharide in water extract and alcohol precipitation groups were 22. 64% and 42. 15%,saponin were 7. 68% and 1. 86%,respectively. Conclusion: 6.0 g/kg Radix Hedysari water extract and alcohol precipitation can enhance anti-oxidation function and the repair of damaged brain tissues,alleviate blood flow changes,regulate the body's immune function,thus reducing levels of brain tissues injury and nerve function disorder induced by acute focal cerebral ischemia. Total polysaccharide and total saponins are the main active substances.
