摘要
目的:观察疏肝泻火方对雌性性早熟大鼠性腺、雌激素和下丘脑促性腺激素释放激素(GnRH)、垂体促性腺激素释放激素受体(GnRH-R)表达的作用。方法:取SD雌性大鼠60只,随机分为正常对照组、性早熟模型组、亮丙瑞林100μg/kg组、疏肝泻火方8.5g/kg、4.25g/kg、2.13g/kg组共6组,采用NMA建立性早熟模型,予不同剂量的疏肝泻火方和亮丙瑞林干预,观察性腺发育情况;测定血FSH、LH水平和下丘脑GnRH、垂体GnRH-R的mRNA表达。结果:性早熟模型组大鼠阴道口开放和出现第一个动情间期的时间分别为32.10±1.56天、35.24±1.46天,均较正常对照组提前,性早熟模型组第一只大鼠出现阴道口开放和第一个动情间期时,疏肝泻火方2.125g/kg组出现一只,其余各组均未见。各组子宫指数和卵巢指数均较模型组显著降低、血清FSH和LH分泌量显著降低,下丘脑GnRH mRNA、垂体GnRH-R mRNA的表达量显著降低,疏肝泻火方8.5g/kg组与亮丙瑞林100μg/kg组疗效相当,疏肝泻火方2.13g/kg、4.25g/kg、8.5g/kg组的作用呈现依次递增的趋势。结论:疏肝泻火方可以降低下丘脑GnRH mRNA和垂体GnRH-R mRNA的表达水平,降低雌激素水平,抑制性腺的发育,延缓HPG轴的启动。
Objective: To observe the effect of Shuganxiehuo recipe on expressions of gonad,estrogen and hypothalamic gonadotropin-releasing hormone(GnRH) and pituitary gonadotropin releasing hormone receptor(GnRH-R) in female precocious puberty rats. Methods: 60 female SD rats were randomly divided into the control group,the precocious model group,100μg/kg leuprorelin group,8. 5 g Shuganxiehuo recipe/kg group,4. 25 g/kg Shuganxiehuo recipe group,2. 125 g/kg Shuganxiehuo recipe group. NMA was used to establish the precocious puberty model. Different doses of Shuganxiehuo recipe and leuprorelin were given. Gonadal development was observed. Levels of serum FSH and LH were determined. mRNA expressions of GnRH in hypothalamus and pituitary GnRH-R were detected. Results: The time of vaginal opening and the first time anoestrum in precocious model group were 32. 10 ± 1. 56 days and 35. 24 ± 1. 46 days,respectively,which happened ahead of those in the control group. When the first vaginal opening and first anoestrum appeared in the precocious model group,which also happened in one mouse of 2. 125 g/kg Shuganxiehuo recipe group,there was no mouse in the other groups. After the model was established,the uterus index and ovarian index increased,serum FSH and LH secretion increased,expressions of GnRH mRNA in hypothalamus and pituitary GnRH-R mRNA increased. Those in the precocious model group were the highest among these groups,and differences were statistically significant compared with precocious model group,(P〈0. 01 or P〈0. 05). The effect of 8. 5 g/kg Shuganxiehuo recipe was equivalent to that of 100μg/kg leuprorelin(P〈0. 05),the effect of Shuganxiehuo recipe was dose-depent. Conclusion: Shuganxiehuo recipe can reduce expression levels of GnRH mRNA in hypothalamus and pituitary GnRH-R mRNA,decrease the level of estrogen,inhibit the development of gonad and delay the activation of HPG axis.
