细菌M19培养条件对降解黄曲霉毒素B_1的影响 被引量:2

摘要 对经筛选的一株降解黄曲霉毒素B_1(AFB_1)的菌株M19进行16S rDNA基因序列分析及系统发育树分析,从而进行菌株鉴定;以AFB_1降解率为评价指标研究培养条件(培养温度、培养时间、接种量、培养基的初始pH、培养基的装液量)对AFB_1降解率的影响。结果表明,该菌株为铜绿假单胞菌(Pseudomonas aeruginosa);培养温度和培养基初始pH对AFB_1降解率的影响较为显著,不同温度及不同初始pH条件下AFB_1降解率存在明显差别(P<0.05),而培养时间、装液量、接种量对AFB_1降解率的影响不显著。培养温度37℃时,AFB_1降解率为90%;初始pH为5时,AFB_1降解率为95.2%。 In this study, 16S rDNA gene sequence analysis and phylogenetic tree analysis were used to identify the strain M19 with degradation activity of AFB1 was isolated and stored in our laboratory. The culture conditions were investigated, including the incubation temperature, incubation time, inoculum size, initial p H of culture medium and medium loading volume, and the degradation efficiency of AFB1 was used as the test index. The results showed that the strain was Pseudomonas aeruginosa, the culture temperature and the initial p H of the medium had significant effects on the degradation rate of AFB1, under the condition of different temperature and initial p H, degradation rate of AFB1 was significantly different(P〈0.05), and the incubation time, loading volume, inoculation size all had no significant effect on AFB1 the degradation rate. When the temperature was 37℃, the degradation rate of AFB1 was 90%, and when the initial p H was 5, the degradation rate of AFB1 was 95.2%.
