摘要
目的 探讨HSP70、HO-1和HSP27在大鼠精索静脉曲张(varicocele,VC)过程中对生精功能的保护作用.方法 选取成年雄性SD大鼠40只随机分为4组,每组10只:假手术组(A组),假手术+槲皮素组(B组),VC组(C组)以及VC+槲皮素组(D组).C组和D组大鼠分别建立精索静脉曲张模型,D组同时腹腔注射槲皮素(100 mg/kg).HE染色观察睾丸组织病理改变;测定丙二醛(MDA)、超氧化物岐化酶(SOD)含量;采用原位缺口末端标记检测细胞凋亡指数;免疫组化检测睾丸组织中HSP70的表达情况;RT-PCR检测HSP27,HO-1 mRNA表达水平.结果 D组严重影响了睾丸结构及生精功能,明显加重了VC引起的组织学损伤;凋亡指数分别为(5.26±0.45)%、(5.83±0.52)%、(14.64±0.98)%、(25.37±1.46)%;SOD活性分别为(0.96±0.06)、(0.88±0.05)、(0.45±0.04)、(0.23±0.03) U/mg蛋白,MDA含量分别为(2.24±0.35)、(2.69±0.32)、(4.78±0.41)、(7.37±0.68) nmol/g蛋白;C组中HSP70阳性表达较A和B组明显增多,D组阳性表达较C组明显减少;与A和B组相比,C组中HSP27、HO-1 mRNA表达水平明显增高.和C组比较,D组中表达水平明显减低.B、C和D三组上述指标相互比较均有统计学意义(P<0.05).结论 HSP70、HO-1和HSP27可减轻氧化应激水平,发挥抗凋亡作用,从而对大鼠精索静脉曲张生精功能产生保护作用.
Objective To investigate the protective effects of HSP70、HO-1 and HSP27 on spermatogenic function in rats with varicocele.Methods Forty adult male SD rats were randomly di vided into 4 groups with 10 rats in each group:sham group (group A),sham + quercetin group (group B),VC group (group C) and VC + quercetin group (group D).An experimental varicocele model was successfully created in group C and D and quercetin(100 mg/kg) was injected intraperitoneally.HE staining was used to observe the pathological changes of testis.Malondialdehyde (MDA) and superoxide dismutase (SOD) content were determined.Cell apoptosis index was detected by dUTP nick end labeling.The expression of HSP70 in testis tissue was detected by immunohistochemistry.The mRNA expression of HSP27 and HO-1 in testis tissue was measured by RT-PCR.Results Testicular structure and spermatogenic function were seriously affected,and histological damage was significantly increased caused by VC in group D.AI was (5.26 ± 0.45) %,(5.83 ± 0.52) %,(14.64 ± 0.98) %,(25.37 ± 1.46) %,respectively.The SOD activities were (0.96 ± 0.06),(0.88 ± 0.05),(0.45 ± 0.04),(0.23 ± 0.03) U/mg protein,MDA content was (2.24 ± 0.35),(2.69 ± 0.32),(4.78 ±0.41),(7.37 ±0.68) nmol/g protein.The expression of HSP70 in group C was significantly higher than that in group A and B,and the expression of HSP70 in group D was significantly lower than that of group C.Compared with group A and B,the mRNA expression level of HSP27 and HO-1 was significantly increased in group C.Compared with group C,the expression level of HSP27 and HO-1 in group D was significantly decreased.The comparison between B、C and D groups was statistically significant (P 〈 0.05).Conclusions HSP70、HO-1 and HSP27 reduces the level of oxidative stress and inhibits the apoptosis.Thus,it plays a protective role on the spermatogenic function in rats with varicocele.
