急性格林巴利综合征患者外周血microRNA-146a、microRNA-125b表达及其与免疫球蛋白、炎症因子的相关性研究 被引量:7

摘要 目的探讨急性格林巴利综合征患者外周血清中microRNA-146a、microRNA-125b的表达水平与免疫球蛋白、炎症细胞因子的相关性。方法选取我院神经内科收治的84例急性格林巴利综合征患者作为试验组(GBS组),另选取50例正常体检者作为对照组。采用RT-PCR技术检测所有纳入者外周血清中microRNA-146a、microRNA-125b的表达水平,采用免疫比浊法检测所有纳入者外周血清中免疫球蛋白G(IgG)、免疫球蛋白A(Ig A)、免疫球蛋白M(Ig M)的水平,采用酶联免疫吸附法检测所有纳入者外周血清中IL-6、CRP、TNF-α的水平。比较2组患者外周血清中microRNA-146a、microRNA-125b的表达水平,采用Spearman相关性分析方法分析microRNA-146a、microRNA-125b分别与免疫球蛋白、炎症因子的相关性。结果试验组外周血清中microRNA-146a表达水平高于正常组,而microRNA-125b的表达水平低于正常组,差异均有统计学意义(P<0.05);试验组外周血清中IL-6、CRP、TNF-α、IgG表达水平高于对照组,差异有统计学意义(P<0.05);Spearman相关性分析表明microRNA-146a与IL-6、CRP、TNF-α、IgG具有正相关性,而microRNA-125b与IL-6、CRP、TNF-α、IgG具有负相关性,差异均有统计学意义(P<0.05);而microRNA-146a及microRNA-125b与Ig A、Ig M无明显相关性。结论急性格林巴利综合征患者外周血清中microRNA-146a、microRNA-125b存在差异表达谱,并且二者可能是通过调控免疫及炎症反应进而参与急性格林巴利综合征的发病。 To investigate the relationship between the expression of microRNA-146a and microRNA-125b and the levels of immune globulin and inflammatory cytokines in peripheral serum of patients with acute Guillain- Barre syndrome, RT-PCR was used to detect the expression of microRNA-146a and microRNA-125b in serum of 84 patients with acute Guillain-Barre syndrome and 50 normal controls. Immunoglobulin assay was used to detect the serum levels of immunolobulin G (IgG), immunoglobulin A (IgA), and immunoglobulin M (IgM) , while enzyme-linked immunosorbent assay (ELISA) was used for the serum levels of IL-6, CRP, and TNF-α in all subjects.The expression levels of microRNA-146a and microRNA-125b in peripheral blood of the two groups werecompared, and Spearman correlation analysis was used to analyze the correlation of microRNA-146a and microRNA-125b with immunoglobulin and inflammatory factors. Data showed that the expression level ofmicrorna-146a in peripheral serum of the experimental group was higher than that of the normal group, while the expression level of microrna-125b was lower than that of the normal group (P〈0.05). The expression levels of IL-6, CRP, TNF-α and IgG in peripheral serum of the experimental group were higher than those of the control group (P〈 0.05). Spearman correlation analysis showed that microRNA-146a was positively correlated with IL-6, CRP, TNF- α, and IgG, while microRNA-125b was negatively correlated with IL-6, CRP, TNF-α, and IgG (P〈0.05). However, microrna-146a and microrna-125b have no obvious correlation with IgA and IgM. In conclusion, there are differential expression profiles of microRNA-146a and microRNA-125b in peripheral serum of patients with acute Guillain-Barre syndrome, and both may participate in the pathogenesis of acute Guillain-Barre syndrome by regulating immune and inflammatory responses.
