细胞外亲环素A与炎性疾病

Extracellular cyclophilin A and inflammatory diseases

亲环素A(cyclophilin A,CyPA)是一种广泛表达的肽基脯氨酰顺反异构酶。在疾病状态下,胞浆中的CyPA可以分泌到细胞外,同时坏死的细胞也会向外释放大量的CyPA,这些分泌/释放到细胞外的CyPA即细胞外CyPA(excellular CyPA,eCyPA)。eCyPA通过与细胞表面受体CD147作用,诱导细胞表达多种炎性细胞因子,并趋化淋巴细胞、单核细胞及粒细胞进入炎症部位,从而在炎性疾病的发生发展过程中发挥重要作用。本文主要介绍近年发现的eCyPA与多种炎症性疾病如类风湿性关节炎、呼吸系统炎症、心血管炎症、牙周炎、胰腺炎、肾炎等发病关系,以冀为这些疾病的诊治提供新的思路。

Cyclophilin A （CyPA） is a widely-expressed peptidyl-prolyl cis-trans isomerase. In disease state, CyPA in the cytoplasm can be secreted to the outside of the cell. At the same time, necrotic cells also release large amounts of CyPA. These secreted/released CyPA outside the cell is called extracellular CyPA （eCyPA）. eCyPA interacts with its receptor CD147 on cell surface, induces cells to express a variety of inflammatory cytokines, and functions as chemockine for lymphocytes, monocytes, and granulocytes to enter the site of inflammation. Thus, eCyPA could play an important role in the development of inflammatory diseases. This review focuses on recent studies on the relationship between eCyPA and inflammatory diseases, such as rheumatoid arthritis, respiratory inflammation, cardiovascular inflammation, periodontitis, pancreatitisand nephritis, to provide new ideas for diagnosis and treatment of these diseases.
