隔药灸对溃疡性结肠炎大鼠结肠黏膜IL-17及IL-23表达的影响 被引量:4

The Influence of Medicine-Separated Moxibustion on Expression of IL-17 and IL-23 in Colonic Mucosa of Rats with Ulcerative Colitis

摘要 目的:观察隔药灸对溃疡性结肠炎(ulcerative colitis,UC)大鼠结肠黏膜中白细胞介素-17(interleukin-17,IL^(-1)7)及白细胞介素-23(interleukin-23,IL-23)表达的影响,探讨隔药灸治疗对UC大鼠Th17细胞因子的调控机制。方法:将44只SD大鼠随机分为正常组12只和造模组32只。采用免疫法加局部刺激造模,将造模成功的30只大鼠随机分为模型组、隔药灸组和西药组,每组各10只。正常组及模型组按照隔药灸组的固定方式固定大鼠,隔药灸组取天枢(双)、气海、中脘等穴位给予隔药灸,西药组灌胃给药柳氮磺胺吡啶。采用免疫组织化学技术法检测各组UC大鼠结肠黏膜中IL^(-1)7、IL-23的表达水平。结果:模型组、隔药灸组和西药组大鼠结肠黏膜中IL^(-1)7、IL-23的水平高于正常组,差异具有统计学意义(P<0.05)。隔药灸组和西药组大鼠结肠黏膜中IL^(-1)7、IL-23水平低于模型组,差异具有统计学意义(P<0.05)。隔药灸组在下调大鼠结肠黏膜中IL^(-1)7、IL-23的表达水平明显优于西药组,差异具有统计学意义(P<0.05)。结论:隔药灸能够调控UC大鼠结肠黏膜中IL^(-1)7、IL-23的表达水平,进而调节UC大鼠结肠黏膜Th17细胞因子平衡。 Objective: To observe the influence of medicine-separated moxibustion on expression of IL-17 and IL-23 in colonic mucosa of rats with ulcerative colitis( UC),and to explore the regulation mechanism of medicine-separated moxibustion on Th17 cytokine in UC rats. Methods: Forty-four SD rats were randomly divided into the normal group with 12 cases and the model-establishing group with 32 cases. Then 30 cases which had been successfully into models were randomly divided into the model group,medicine-separated moxibustion group and Western medicine group,with 10 rats in each group. In the normal group and the model group,the rats were fixed in accordance with the fixed mode of the medicine-separated moxibustion group. The medicine-separated moxibustion group was given medicine-separated moxibustion on double Tianshu( ST 25),Qihai( CV 6),Zhongwan( CV 12) and other acupoints. The Western medicine group was given sulfasalazine for gavage. Immunohistochemical method was used to detect the expression of IL-17 and IL-23 in colon mucosa of UC rats in each group. Results: The levels of IL-17 and IL-23 in the colon mucosa of the model group,the medicine-separated moxibustion group and the Western medicine group were higher than those of the normal group( P 〈0. 05). The levels of IL-17 and IL-23 in colon mucosa of rats in the medicine-separated moxibustion group and the Western medicine group were lower than those of the model group,and the differences were statistically significant( P〈 0. 05). The expression levels of IL-17 and IL-23 in colon mucosa of rats were significantly better than that of Western medicine group,and the difference was statistically significant( P 〈0. 05). Conclusion: Medicine-separated moxibustion can regulate the expression levels of IL-17 and IL-23 in the colonic mucosa of UC rats,and then regulate the balance of Th17 cytokines in them.
